The enhancement of differentiation inductors effects on the solid tumors model in vitro

Abstract:

Indukovaná diferenciace nádorově transformovaných buněk do terminálních stádií představuje v současné době jednu ze slibných strategií protinádorové terapie. V posledních letech se pozornost zaměřuje také na možnosti kombinované indukce diferenciace, kdy je diferenciační efekt jednoho činidla modulován souběžnou aplikací dalších sloučenin. Tento terapeutický přístup byl úspěšně demonstrován nejen na in vitro modelech, ale rovněž v klinické praxi především u hematologických malignit. Mezi velmi často používané induktory diferenciace patří retinoidy, přičemž jimi indukovaná diferenciace může být ještě dále potencována např. inhibitory takových enzymů, které participují na intracelulární degradaci retinoidů; jedná se zejména o inhibitory lipoxygenáz (LOX) a cyklooxygenáz (COX). Předmětem předkládané disertační práce bylo studium možných mechanismů vlivu kombinované aplikace LOX/COX inhibitorů společně s kyselinou all-trans retinovou (ATRA) na modelu stabilizovaných buněčných linií, které byly derivovány z neuroblastomu - linie SK-N-BE(2) a SH-SY5Y, a z meduloblastomu – linie Daoy a D283. Jako modulátory biologického efektu ATRA byly použity kyselina kávová (caffeic acid, CA) coby inhibitor 5-lipoxygenázy a celecoxib (CX) inhibující cyklooxygenázu 2. U obou buněčných linií derivovaných z neuroblastomu byla schopnost indukce diferenciace směrem k neuronálnímu fenotypu potvrzena již po aplikaci samotné ATRA. Zesílení diferenciačního efektu ATRA bylo patrné zejména po společné aplikaci s CA, a to u obou studovaných neuroblastomových linií. V případě aplikace CX - a to ať už samotného, nebo v kombinaci s ATRA - ovšem u obou linií predominoval spíše cytotoxický efekt. Všechny provedené analýzy rovněž potvrdily vyšší citlivost buněk linie SK-N-BE(2) vůči kombinovanému působení ATRA a LOX/COX inhibitorů, což koresponduje s poznatky o biologických vlastnostech této linie, kdy buňky SK-N-BE(2) byly popsány jako nediferencované, odpovídající spíše fenotypu nádorových kmenových buněk. U buněčných linií Daoy a D283 derivovaných z meduloblastomu nebyla na morfologické úrovni potvrzena schopnost ATRA indukovat neuronální diferenciaci, avšak z výsledků proliferačních studií a flow-cytometrické analýzy obsahu DNA byl patrný potenciál ATRA indukovat apoptózu. Jako citlivější se k působení ATRA a/nebo LOX/COX inhibitorů jevila linie D283. Důvodem takových rozdílů v odpovědi na aplikované experimentální zásahy byl pravděpodobně odlišný typ buněk, který tyto linie reprezentují – zatímco u buněk linie D283 byl prokázán jejich neuronální původ, u buněk linie Daoy se jednalo o smíšený neuro-gliální původ, který buňkám propůjčuje vyšší odolnost vůči induktorům diferenciace typu ATRA. Ačkoli výsledky ukazují rozdílnou senzitivitu použitých neuroblastomových a meduloblastomových linií ke zvoleným experimentálním zásahům, lze shrnout, že kombinovanou aplikací ATRA společně s LOX/COX inhibitory může být antineoplastický potenciál ATRA dále zesilován.

Abstract:

Induced differentiation of transformed cells into mature phenotypes is considered to be one of the most promising strategies of recent antitumor therapy. The effects of differentiation inductors may be further potentiated by their co-application with other compounds. This therapeutic approach heas been successfully demonstrated both on tumor cell lines and in clinical practice, especially in treatment of hematological malignancies. Retinoids represent the frequently used inductors of cell differentiation and their effects may be influenced by inhibitors of enzymes participating in the intracellular degradation pathway of retinoids, i.e. especially of lipoxygenases (LOX) and cyclooxygenases (COX). This work was aimed at a detailed study of the effects of combined application of all-trans retinoic acid (ATRA) with the LOX/COX inhibitors in tumor cells of selected types of pediatric tumors. Established cell lines derived from neuroblastomas (SK-N-BE(2) and SH-SY5Y) and medulloblastomas (Daoy and D283) were chosen as biological models. Caffeic acid (CA) as inhibitor of 5-lipoxygenase and celecoxib (CX) as inhibitor of cyclooxygenase 2 were used in this study. The capability of ATRA to induce cell differentiation into neuronal phenotype was confirmed in both neuroblastoma cell lines. The enhancement of this differentiation effect was evident especially after combined application of ATRA and CA. In contrast, CX showed cytotoxic effect predominantly, whether it was applied either alone or in combination with ATRA. All the employed analyses also confirmed higher sensitivity of SK-N-BE(2) cells to applied treatment. These findings are in accordance with recently published data that describe SK-N-BE(2) cell line as non-defferentiated cells exhibiting tumor stem cell phenotype. In medulloblastoma cell lines the potential of ATRA to induce neuronal differentiation has not been proven according to the absence of changes in cell morphology. On the contrary, cell proliferation assays and flow-cytometric analysis of DNA content revealed the potential of ATRA to induce apoptosis in both medulloblastoma cell lines. D283 cell line was found to be more sensitive to ATRA and/or LOX/COX inhibitors. The dissimilarities in response of both medulloblastoma cell lines to the applied treatment were probably caused by different cell type – D283 cells are of neuronal origin while Daoy cells are of mixed neuro-glial origin which impart these cells partial resistance to induced cell differentiation. Although the results show some differences in sensitivity of studied cell lines to applied LOX/COX inhibitors and also distinct biological effects of their combined application with ATRA, it can be concluded that the antineoplastic effects of ATRA may be potentiated by the combined application of ATRA with LOX/COX inhibitors.
