The combination of separation techniques and mass spectrometry for the proteomic analysis of barley

Abstract:

Ječmen je důležitou zemědělskou plodinou používanou k přípravě sladu, jedné z výchozích surovin při výrobě piva. Chemické složení ječmene tak ovlivňuje výsledné vlastnosti tohoto hospodářsky významného nápoje. Jedním ze sledovaných parametrů ječmene je jeho proteinový obsah. Tato diplomová práce se zabývá charakterizací a porovnáním obsahu vodorozpustných proteinů obsažených v zrnu a sladu ječné sladovnické odrůdy Jersey. K tomu byla využita kombinace 2D gelové elektroforézy, enzymatického štěpení vybraných proteinů a jejich identifikace pomocí MALDI-TOF MS a MS/MS analýz a následného databázového vyhledávání. Jedním z cílů byla také detekce proteinů modifikovaných glykosylací, která významně ovlivňuje vlastnosti proteinů. K dosažení tohoto cíle bylo použito afinitní chromatografie následované SDS-PAGE, enzymovým štěpením a MALDI-TOF MS a MS/MS analýzou. Použitím metody zahrnující dvourozměrnou gelovou elektroforézu byly identifikovány jak proteiny přežívající sladování (např. glyceraldehyd-3-fosfát dehydrogenasa) tak i proteinové spoty specifické pro zrno (B3-hordein) či slad (Barperm1, ječný hypotetický protein, atd. ). Lektinová afinitní chromatografie následovaná jednorozměrnou elektroforézou odhalila dva potenciální proteiny modifikované glykosylací, jeden v zrnu (protein disulfid-isomerasa) a jeden ve sladu (serin-karboxypeptidasa). Navíc byla identifikována i řada významných ječných proteinů, které nebyly lektinovou AC zadržovány (protein Z, beta-amylasa, endochitinasa, atd.).

Abstract:

Barley is an important agricultural crop mainly used for the production of malt, one of the key ingredients for brewing beer. Therefore, the chemical composition of barley affects the resulting properties of beer. Barley protein composition is one of widely studied aspects, which is also the focus of this master's thesis. The goals of this thesis were to characterize and compare water-soluble proteins found in grain and malt, and to detect and identify proteins modified by glycosylation, which gives proteins unique properties. To achieve these goals, the combination of two-dimensional gel electrophoresis, one-dimensional gel electrophoresis and affinity chromatography were employed as the separation methods. The identification and characterization of selected proteins was carried out by MALDI-TOF MS and MS/MS analyses followed by database searching. The use of two-dimensional gel electrophoresis led to the identification of both proteins surviving the malting process (e.g. glyceraldehyde-3-phosphate dehydrogenase) and protein spots that were specific for grain (B3-hordein) or malt (Barperm1, barley hypothetical protein, etc.). Affinity chromatography followed by one-dimensional gel electrophoresis revealed two potential N-glycoproteins, one found in grain (protein disulfide-isomerase) and one found in malt (serine carboxypeptidase). Moreover, many important barley proteins were identified in lectin non-bound fractions (protein Z, beta amylase, endochitinase, etc.).
