Distribution of free amino acids and biogenic amines content in the eidam type cheese

Abstract:

Cílem této diplomové práce bylo sledování distribuce volných aminokyselin a biogenních aminů v sýrech holandského typu během jejich zrání. Sýry zrály při teplotách 10 ? 2 °C a 16 ? 2 °C. U všech vzorků byla provedena základní chemická analýza (pH, obsah sušiny a NaCl). Obsahy vybraných biogenních aminů i volných aminokyselin byly stanoveny pomocí iontově ? výměnné kapalinové chromatografie s fotometrickou detekcí. U sýrů zrajících při vyšší teplotě bylo zpozorováno, že dochází k rychlejšímu uvolňování volných aminokyselin z proteinů, v 56. dnu bylo toto množství téměř dvojnásobné než u kontrolních vzorků. Koncentrace biogenních aminů a volných aminokyselin byla sledována ve třech vrstvách třech plátů. Největší množství uvolněných aminokyselin byla zpozorována v jádře, a to u obou sýrů. Obsahy biogenních aminů byly také vyšší u sýrů zrajících při vyšší teplotě. V 56. den dosahoval obsah tyraminu u některých vrstev sýrů zrající při vyšší teplotě hodnot převyšujících 400 mg.kg-1.

Abstract:

The aim of this thesis was to monitoring distribution of the free amino acids and biogenic amines in the Dutch ? type cheese. This cheese had matured at 10 ? 2 °C and 16 ? 2 °C. All samples were subjected to basic chemical analysis (pH, content of dry matter and NaCl). The content of biogenic amines and free amino acids was determined by using ion ? exchange liquid chromatography with spectrophotometric detection. The cheese which was ripening by higher temperature contained greater amount of free amino acids, by 56th day was amount of free amino acid almost doubly greater than in control cheese. The amount of biogenic amines and free amino acids was detected in three layers of each of three plates. The greater amount of free amino acids was detected in centre of each plate. Also amount of biogenic amines were detected in cheese which where stored in 16 ? 2 °C. In the 56th day, the tyramine content in some layers of accelerated cheese reached the values above 400 mg.kg-1.
