Abstract:

Tato diplomová práce pojednává o využití hydroxyskořicových kyselin k inhibici malolaktické fermentace u červených vín. Teoretická část je hlavně zaměřena na hydroxyskořicové kyseliny, a to konkrétně na kávovou, ferulovou a kumarovou, a také na kyselinu jantarovou a použité bakterie mléčného kvašení. Experimentální část je zaměřena na tvorbu růstových křivek a diskové difúzní metody pro každou bakterii (Oenococcus oeni, Lactobacillus brevis a Lactobacillus plantarum) a použitý inhibitor (kyselinu). Je prokázáno, že všechny inhibitory ve vyšších koncentracích mají inhibiční účinky na dané bakterie mléčného kvašení. Inhibiční účinek je velmi podobný pro všechny tři bakterie. V další části experimentu jsou provedeny inhibiční pokusy na červeném víně s koncentracemi inhibitorů 0,25; 0,5 a 1 g·l−1. Vzorky jsou podrobeny analýze pomocí HPLC (vysokoúčinná kapalinová chromatografie) ke stanovení koncentrací kyseliny jablečné a mléčné. Nejvyšší inhibiční účinky má kyselina ferulová a kumarová při koncentraci 1 g·l−1, při nejnižší koncentraci 0,25 g·l−1 se nejlépe osvědčila kyselina kávová.

Abstract:

This thesis deals with the use of hydroxycinnamic acids to inhibit of malolactic fermentation in red wines. The theoretical part is mainly focused on hydroxycinnamic acids, namely caffeic, ferulic and coumaric acids, as well as succinic acid and the lactic acid bacteria. The experimental part is focused on the creation of growth lines and disk diffusion methods for each bacteria (Oenococcus oeni, Lactobacillus brevis a Lactobacillus plantarum) and the used inhibitor (acid). All inhibitors at higher concentrations have been shown to have inhibitory effects on the lactic acid bacteria. The inhibitory effect was very similar for all three bacteria. In the next part of the experiment, inhibition experiments are performed on red wine with inhibitor concentrations of 0.25; 0.5 and 1 g·l−1. The samples underwent analysis through HPLC (high-performance liquid chromatography) to determine the concentrations of malic and lactic acid. Ferulic and coumaric acid have the highest inhibitory effects at a concentration of 1 g·l−1, while caffeic acid proved to be the best at the lowest concentration of 0.25 g·l−1.
