Possibilities of using probiotic cultures in beer production

Abstract:

V této diplomové práci bylo pojednáno o potenciálu probiotických kultur ve vztahu k lidskému zdraví a jejich výskytu ve fermentovaných potravinách včetně piva a byly popsány limitující faktory pro výrobu probiotického piva. Blíže byly specifikovány ně-které bakterie mléčného kvašení rodu Lactobacillus a Streptococcus a také bakterie rodu Bifidobacterium a kvasinka Saccharomyces boulardii, které byly použity v experimentální části práce. Do mladiny o 10 % EPM byly přidány v různých fázích fermentace probiotické kultury a bylo prováděno měření pH, alkoholu a skutečného ex-traktu v průběhu fermentace. Cílem senzorické analýzy bylo zjistit, zda se použití kon-krétní kultury projeví na senzorickém profilu daného piva. Ke změnám v senzorickém profilu oproti kontrole bez probiotické kultury sice u většiny vzorků docházelo, ale ne ve významné míře. Mírně větší vliv na senzorický profil byl pozorován v závislosti na pří-davku kultury do různých fází fermentace piva. Barvením buněk methylenovou modří byla sledována viabilita kvasinek a po součtu barvených a nebarvených buněk, byla sta-novena viabilita, která byla u kvasinky S. boulardii po 32 dnech 87, 63 %. Tato práce může čtenáři posloužit při výběru kultury a nastavení podmínek fermentace pro výrobu piva s přídavkem probiotických kultur.

Abstract:

This diploma thesis deals with potential of probiotic cultures in relation to human health and occurrence of these cultures in fermented foods, including beer, was discussed. Also limiting factors for the production of probiotic beer were described. Some lactic acid bacteria of the genus Lactobacillus and Streptococcus were specified, as well as bacteria of the genus Bifidobacterium and the yeast Saccharomyces boulardii, which were used in the experimental part of the work. Probiotic cultures were added to the wort at 10 % EPM at various stages of fermentation and pH, alcohol and real extract were measured during the fermentation. The aim of sensory analysis was to determine whether the use of a particular culture is reflected in the sensory profile of a given beer. Although the changes in the sensory profile compared to the control without probiotic culture occurred as not significant in most of the samples. Slightly greater effect on the sensory profile was observed depending on the addition of culture to different phases of beer fermentation. The viability of the cells was determined by staining the cells with methylene blue, and after summing the stained and unstained cells, the viability was determined. The yeast cell viability of S. boulardii was 87.63% after 32 days of fermentation. This thesis can help the reader in selecting the culture and setting the fermentation conditions for beer production with the addition of probiotic cultures.
