Detection of Mycoplasma hominis and Ureaplasma urealyticum by real-time PCR

Abstract:

Tato diplomová práce se zabývá laboratorním průkazem Mycoplasma hominis a Ureaplasma urealyticum v genitálním traktu žen z gynekologické ambulance FN Hradec Králové a mužů z Centra asistované reprodukce SANUS Pardubice. Průkaz uvedených druhů byl prováděn druhově specifickou SYBR green PCR metodou, která byla zavedena a optimalizována v průběhu řešení diplomové práce. Celkově bylo vyšetřeno 27 stěrů z děložního čípku a 18 ejakulátů. M. hominis jsme prokázáli ve 3 (11,1 %) stěrech a U.urealyticum v 9 (33,3 %) stěrech. Směsný výskyt jsme zaznamenali ve 2 (7,4 %) stěrech a 1 ejakulátu (5,5 %) Výskyt U. urealyticum jsme zaznamenali s vyšší frekvencí, a to zejména u žen.

Abstract:

The aim of this thesis is focused on the laboratory detection Mycoplasma hominis and Ureaplasma urealyticum in the urogenital tract of women from gynecological clinic of University Hospital Hradec Králové and men from the Centre of Assisted Reproduction SANUS Pardubice. Detection of these species was carried out species-specific SYBR green PCR method which we have implemented and optimized. Overall, we examined 27 cervical swabs and 18 ejaculates. M. hominis was detected in 3 (11,1 %) women and U. urealyticum was detected in 9 (33,3 %) women. The incidence of both pathogens in the mixture we detected in 2 (7,4 %) cervical swabs and in 1 (5,5 %) ejaculate. The incidence of U. urealyticum was registred with a higher frequency, especially in women.
