Abstract:

Mouse lymphoma assay je in vitro toxikologický test využívající k testování potenciálního genotoxického účinku chemických látek savčích buněk. Těmito buňkami jsou buňky myší lymfomatické buněčné linie L5178Y/Tk+/- -3.7.2.C. V důsledku působení toxické látky dochází ke ztrátě alely pro tymidinkinázu a tím i schopnosti ji produkovat. Takové mutantní buňky jsou pomocí pyrimidinového analogu trifluortymidinu selektovány a na základě jejich růstu se posléze určuje míra toxicity látky. Testovanými látkami byly: kyselina mukobromová, formaldehyd a cyklofosfamid a kolchicin jako pozitivní kontrola. Metoda MLA byla porovnána z hlediska náročnosti na přípravu, časové náročnosti a náchylnosti ke kontaminaci a v důsledku toho i ke zkreslení výsledků s metodami jiných testů určených k testování možné genotoxicity chemických látek, jako jsou in vitro mikronukleus test. V provedeném in vitro mikronukleus testu vykázala koncentrace formaldehydu 0,02 ?l/ml pozitivní výsledek, tj. dvojnásobné zvýšení výskytu aberací oproti negativní kontrole bez MAS. Vyšší koncentrace formaldehydu byly silně cytotoxické. Předpokládaná toxicit kyseliny mukobromové nebyla prokázána.

Abstract:

Mouse lymphoma assay is in vitro toxicology test that uses mammalian cells for testing potential genotoxic effects of chemicals. In this test are used of L5178Y/Tk+/- -3.7.2.C mouse lymphoma cell line. The toxic substances caused defection of thymidin kinase allele and cells have not ability to produce it. These mutant cells are selected by pyrimidine analog trifluorothymidine and on the basis of the cell growth rate is than finally identified a level of the toxicity of the substance. The tested chemical substances were: mucobromic acid, formaldehyde and cyklophosphamide with colchicine as positive controls. Method of mouse lymphoma assay was confronted with methods of other test of genotoxicity like in vitro mammalian cell micronucleus test in light of heftiness on disposition, time disposition and disposition to contamination that caused distortion results. In in vitro mammalian cell micronucleus test was formaldehyde concetration 0,02 ?l/ml positive i. e. formaldehyde induced significant increase of chromosomal aberation compared with negative control without MAS. Higher formaldehyde concetrations were cytotoxic. Mucobromic acid was negative in in vitro mammalian cell micronucleus test.
