Study of reactivation of selected p53 mutants in B-cell tumor cell lines

Abstract:

Mutations and/or deletions in the TP53 gene occur in approximately 10 to 15 % of patients with haematological malignancies. Their presence leads to a very poor prognosis, which includes the need for therapy and - despite it - frequently results in the short survival. The subject of diploma thesis was to study the reactivation of selected p53 mutants in tumor cell lines (RAJI, RAMOS, SU-DHL-4) using commercially available experimental agents (PRIMA-1, ellipticine). These small molecules are able to alter the conformation of mutated p53 protein to its native conformation. It was assumed that - in the case of successful reactivation - the standard functions of the p53 protein will result into reduction of the p53 protein level and subsequent induction of the p53 target genes, i.e., PUMA, BAX, and CDKN1A. Therefore, the thesis methodically included the work with tissue cultures (leukaemia and lymphoma cell lines), including their treatment with reactivating substances, analysis of cell metabolic activity (WST-1 assay), western blotting, immunoprecipitation and quantitative real-time PCR. Our results show that the small molecules - PRIMA-1 and ellipticine - led in some cases to (1) the concentration-dependent reduction of mutant p53 protein level, and (2) to the concentration-dependent induction of CDKN1A and BAX genes regulated by the p53 protein. This most likely demonstrates the ability of these small molecules to reactivate the selected p53 mutants. The effects of these molecules on tumor cells with other p53 mutations will be a subject of the follow-up study, which will also include an evaluation of their effect in combination with chemotherapeutic agents.

Abstract:

Mutácie a/alebo delécie v géne TP53 sa objavujú u asi 10 – 15 % pacientov s hematologickými malignitami. Ich prítomnosť vedie k veľmi zlej prognóze, takmer vždy vyžadujúcej nevyhnutnosť terapie a napriek tomu k veľmi krátkej dobe prežívania. Predmetom diplomovej práce je štúdium reaktivácie vybraných p53 mutantov v bunkách nádorových línií (RAJI, RAMOS, SU-DHL-4) prostredníctvom komerčne dostupných experimentálnych látok (PRIMA-1, elipticín) schopných meniť mutovanú konformáciu proteínu p53 na funkčnú. Predpokladá sa, že v prípade úspešnej obnovy štandardnej funkcie mutovaného proteínu dôjde k zníženiu hladiny p53 v bunkách a následne k indukcii expresie cieľových génov p53, napríklad PUMA, BAX, a CDKN1A. Diplomová práca preto metodicky zahŕňala najmä prácu s tkanivovými kultúrami leukemických a lymfómových buniek, vrátane ich ošetrovania testovanými látkami, analýzy metabolickej aktivity (WST-1), westernový prenos, imunoprecipitáciu a kvantitatívnu polymerázovú reťazovú reakciu v reálnom čase. Vo výsledku „malé molekuly“ (PRIMA-1, elipticín) viedli v niektorých prípadoch ku koncentračne závislému zníženiu hladiny mutovaného proteínu p53. V iných prípadoch zase došlo ku koncentračne závislej indukcii génov CDKN1A a BAX, teda génov regulovaných proteínom p53. Toto s veľkou pravdepodobnosťou preukazuje schopnosť týchto molekúl reaktivovať vybraných p53 mutantov. Účinky týchto molekúl na nádorové bunky s ďalšími mutáciami proteínu p53 budú predmetom nadväzujúceho štúdia, rovnako ako hodnotenie ich účinku v kombinácii s chemoterapeutikami.
