Endocrine discruptive potential of complex environmental samples

Abstract:

V poslední době jsou stále více studovány negativní účinky cizorodých látek, přítomných v životním prostředí, na lidi a volně žijící organizmy. Vyvstává tak potřeba vyvinout vhodné metody pro stanovení míry znečištění prostředí. Chemická analýza pro tento účel plně nevyhovuje, protože v prostředí se vyskytují tisíce jednotlivých látek, pro jejichž stanovení zatím neexistují vhodné analytické metody. Navíc, samotná chemická analýza není schopna podat informace o toxicitě jak jednotlivých chemikálií, tak jejich komplexních směsí, v kterých se obvykle v prostředí vyskytují. Kromě nespecifické toxicity, může řada kontaminantů způsobovat specifické efekty, jako je endokrinní disrupce nebo toxicita dioxinového typu, které je činí zvláště toxikologicky významnými. Proto je třeba vyvinout nové metody zaměřené konkrétně na tyto specifické mechanizmy účinku. Předkládaná práce popisuje použití in vitro metod pro hodnocení specifických mechanizmů účinku komplexních extraktů ze sedimentů, půdy nebo vzduchu a modelových látek, které reprezentují některé skupiny polutantů. Použité metody zahrnují stanovení „klasických“ specifických efektů jako toxicity dioxinového typu nebo estrogenity, ale i méně prozkoumaných mechanizmů účinku, jako je ovlivnění aktivity retinoidní signální dráhy nebo produkce steroidů. Předkládaná práce ukazuje užitečnost in vitro metod pro hodnocení úrovně znečištění životního prostředí s ohledem na specifické efekty vyvolané přítomnými polutanty. Tyto metody poskytují dodatečné informace k datům z chemických analýz. Předkládané výsledky naznačují význam in vitro metod v biomonitoringu pro správné zhodnocení rizik.

Abstract:

There is increasing concern over the potential adverse effects of xenobiotics present in the environment on human and wildlife populations and so new methods to assess the level of pollution in the environment are needed. Chemical analyses cannot quantify the levels of all pollutants complexly because there are thousands of individual chemicals present in environmental matrices. Moreover, the analyses do not describe the toxicity of the pollutants or their complex mixtures. Besides nonspecific toxicity, some of the contaminants possess a specific toxic mode of action such as dioxin-like toxicity or endocrine disruptive potential that makes them even more dangerous. Thus, it is necessary to develop bioassays specifically addressing these modes of action. This work describes a series of studies dealing with application of in vitro bioassays in evaluation of specific toxicity of complex extracts from different environmental matrices, such as air, soil and sediment, and model compounds representing several classes of pollutants. The used methods include both ‘classical’ specific toxic effects such as dioxin-like toxicity, estrogenicity, androgenicity and relatively novel modes of action such as potential to interfere with retinoid signaling or production of steroids. The present work shows utility of in vitro bioassays in evaluation of the level of environmental pollution with respect to specific effects of present contaminants. The bioassays provide additional information to data from chemical analyses. The results emphasize the important role bioassays can play in environmental biomonitoring for the correct risk assessment and the complementarity of bioassay results with data from chemical analysis.
