Ancient DNA quantification of the skeletal remains from the burial site Diváky - Padělky za humny

Abstract:

Z kosterních pozůstatků z pohřebiště Diváky – Padělky za humny byla izolována a následně kvantifikována archaická DNA. Z každého jedince byly odebrány zub a fragment z kosti za účelem porovnání výtěžnosti aDNA. Dále byl každý vzorek izolován pomocí MinElute® PCR Purification Kit firmy QIAGEN, PrepFiler® Forensic DNA Extraction Kit firmy Life Technologies a pomocí nově zaváděné extrakce DNA s alkoholovou precipitací, opět za účelem porovnání úspěšnosti jednotlivých metod. Kvantifikace byla prováděna pomocí UV spektrofotometru firmy Boeco a Plexor® HY System firmy Promega. Jednotlivé koncentrace aDNA byly navzájem porovnávány za účelem vyhodnocení nejúspěšnější metody a zdroje archaické DNA. Ovšem na 95% hladině významnosti nebyl ani v jednom případě prokázán statistický rozdíl, zřejmě z důvodu malého souboru testovaných dat. Úspěchem bylo zjištění, že nově zaváděná metoda DNA extrakce s alkoholovou precipitací je podobně spolehlivá jako dvě komerční metody a proto je možné ji v budoucnosti používat v Laboratoři biologické a molekulární antropologie Ústavu experimentální biologie Přírodovědecké fakulty Masarykovy univerzity.

Abstract:

Ancient DNA from the skeletal remains from the burial site Diváky – Padělky za humny was extracted and quantificated. There were ten graves analyzed, from each was obtained a tooth and a bone fragment to compare the quantity of ancient DNA. Furthermore each sample was extracted by MinElute® PCR Purification Kit by the company QIAGEN, PrepFiler® Forensic DNA Extraction Kit by the company Life Technologies and finally by a new method based on DNA extraction and ethanol precipitation introduced to LBMA. Quantification was made by the UV spectrophotometry and the Plexor® HY System (Promega). The purpose of extraction and quantification was to compare efficiency of each method and the origin of a skeleton and to obtain knowledge about the best method for the ancient DNA use. However, there was no statistical meaningful difference between each group of samples, probably because of the low sample amount. The success of the thesis was the establishment that the new method is comparably successful as the other ones and it is possible to use it in the future in the LBMA.
