Gene TP53 And Li Fraumeni Syndrome

Abstract:

Tato bakalářská práce je věnována problematice genu TP53 a Li-Fraumeniho syndromu, který je způsoben zárodečnými mutacemi v tomto genu. Produktem genu TP53 je protein p53, jehož funkce spočívá v ochraně organismu proti vzniku nádorových onemocnění. Je schopen navodit apoptózu neboli řízenou buněčnou smrt buněk, pozastavit buněčný cyklus a poskytnout tak reparačním enzymům čas potřebný na opravu poškozené DNA. V případě, že dojde k mutaci genu TP53, sníží se tím pádem i ochrana proti vzniku nádorových onemocnění. Onemocnění, které je způsobeno mutacemi tohoto genu, je Li-Fraumeni syndrom. Pacienti s tímto onemocněním mají vysokou pravděpodobnost vzniku nejrůznějších nádorových onemocnění již v dětském věku.

Abstract:

This bachelor thesis is devoted to gene TP53 and Li-Fraumeni syndrome, which is caused by germline mutations in this gene. The product of the TP53 gene is the p53 protein, whose function is to protect the body against cancer illnesses. It is able to induce apoptosis or controlled cell death, to suspend the cell cycle and to provide the time needed for repair enzymes to repair demaged DNA. If the gene TP53 is mutated, the protection against cancer illnesses is reduced. The disease caused by mutations in this gene is called Li-Fraumeni syndrome. Patients with this disease have high probability of developing various cancers already in childhood.
