Application of cytogenetic methods in protection of public health

Abstract:

Diplomová práce přímo navazuje na autorčinu bakalářskou práci zaměřenou na cytogenetickou analýzu lidských periferních lymfocytů (CAPL) a rozšiřuje zaměření na aplikaci aktuálních cytogenetických metod v ochraně veřejného zdraví, které představují důležitý nástroj při sledování vlivu genotoxických látek na organismus a také monitoring expozice genotoxickým látkám v komunálním i pracovním prostředí. Biologickým projevem těchto faktorů, které působí na organismus, mohou být chromozomové změny a abnormality. Práce se zaměřuje na nejvýznamnější cytogenetické metody a zhotovena byla formou rešerší odborné literatury. V praktické části autorka hodnotila pomocí cytogenetické analýzy a Mikronukleus testu probandy exponované genotoxickým faktorům v pracovním prostředí. Autorčiny výsledky hodnocení a procentuální aberace byly porovnány s výskytem spontánních aberací v populaci. Praktická část byla také zaměřena na nejnovější studie zabývající se problematikou expozice genotoxickým faktorům v pracovním prostředí a na základě těchto studií autorka provedla meta-analýzu. Předpokladem autorčina hodnocení bylo to, že z důvodu expozice probandů genotoxickým látkám v zaměstnání bude výskyt chromozomálních aberací zvýšen nad hodnotu spontánních aberací v populaci.

Abstract:

The diploma thesis directly follows the author's bachelor's thesis focused on the cytogenetic analysis of human peripheral lymphocytes (CAPL) and expands the focus on the application of current cytogenetic methods in public health protection, which represent an important tool for monitoring the influence of genotoxic substances on the organism and also monitoring exposure to genotoxic substances in municipal and the work environment. Chromosome changes and abnormalities can be a biological manifestation of these factors that affect the organism. The thesis focuses on the most important cytogenetic methods and was made in the form of a search of professional literature. In the practical part, the author evaluated probands exposed to genotoxic factors in the work environment using cytogenetic analysis and the Micronucleus test. The author's evaluation results and percentage aberrations were compared with the incidence of spontaneous aberrations in the population. The practical part was also focused on the latest studies dealing with the issue of exposure to genotoxic factors in the work environment, and based on these studies, the author performed a meta-analysis. The assumption of the author's assessment was that due to the exposure of the probands to genotoxic substances at work, the occurrence of chromosomal aberrations will be increased above the value of spontaneous aberrations in the population.
