Evaluation of DNA repair gene expression during oxidative stress

Abstract:

Oxidační stres je v dnešní době velmi reflektované téma. Je to hlavně z důvodu, že zvýšená hladina oxidačních činidel, která je za tento stav zodpovědná, způsobuje poškození makromolekul a dalších buněčných struktur. Látky, které zabraňují vzniku tohoto stavu, či jej eliminují, jsou označovány jako antioxidanty. V případě, kdy se nezdaří zabránit vzrůstu koncentrace oxidačních činidel nebo nenastane jejich eliminace, dochází velmi často k poškození buňky. V tomto případě nastupují antioxidační reparační mechanismy. Ve své bakalářské práci jsem se zaměřila na oxidační stres na úrovni DNA. V rámci teoretické části bylo cílem vyhledat lidské geny, které se podílejí na opravě DNA poškozené působením oxidačního stresu a fungují tedy jako antioxidační reparační systém molekuly DNA. Součástí teoretické části je i popis metodiky, kterou lze expresi DNA opravných genů sledovat a hodnotit. V praktické části mojí práce bylo cílem popsanou metodiku z teoretické části vyzkoušet experimentálně a také sledovat a hodnotit změny v expresi DNA opravných genů při působení peroxidu vodíku, který je zodpovědný za oxidační stres v buňce. Hypotéza pro tento experiment zní, že geny, které se podílejí na opravě DNA poškozené peroxidem vodíku, zvýší svou expresi v jeho přítomnosti.

Abstract:

Nowadays, oxidative stress is a very reflective topic. This is mainly because the increased level of oxidizing agents responsible for this condition causes damage to macromolecules and other cellular structures. Substances that prevent or eliminate this condition are referred to as antioxidants. If the concentration of oxidizing agents is not reduced or eliminated, cell damage is very common. In this case, antioxidant repair mechanisms begin to work. In my bachelor thesis I focused on oxidative stress at the DNA level. In the theoretical part, the aim was to find human genes involved in the repair of DNA damaged by oxidative stress and thus function as an antioxidant DNA repair system. The theoretical part also includes a description of the methodology by which the expression of DNA repair genes can be monitored and evaluated. In the practical part of my work the aim was to attempt the described methodology from the theoretical part experimentally and also to evaluate and monitor changes in the expression of DNA repair genes under the influence of hydrogen peroxide, which is responsible for oxidative stress in the cell. The hypothesis for this experiment is that the genes involved in repairing DNA damaged by hydrogen peroxide will increase their expression in its presence.
