Determination of dissociation constant of some drugs using spectrophotomentric and potentiometric titration data

Abstract:

Metodou potenciometrické titrace byla stanovena termodynamická disociační konstanta léčiva hydroxyurea při teplotách 25°C a 37°C. Regresní analýzou naměřených dat pomocí programů ESAB a HYPERQUAD byly na základě nejlepší těsnosti proložení určeny smíšené disociační konstanty pKa. Ze závislosti stanovených smíšených disociačních konstant na odmocnině z iontové síly při teplotách 25°C a 37°C byly regresní analýzou Debye-Hückelova vztahu ve statistickém programu ADSTAT získány termodynamické disociační konstanty pKaT. Metodou spektrofotometrické titrace s UV detekcí byly stanoveny termodynamické disociační konstanty léčiv alendronátu sodného a sildenafil citrátu; měření bylo provedeno při teplotě 25°C a 37°C. Pomocí regresní analýzy absorbanční responzní plochy programem SQUAD(84) a SPECFIT32 byly stanoveny smíšené disociační konstanty pKa. Ze závislosti těchto smíšených disociačních konstant na odmocnině z iontové síly při dané teplotě byla regresní analýzou Debye-Hückelova vztahu určena termodynamická disociační konstanta pKaT. Chemický model pro dané léčivo a počet světlo-absorbujících částic byl určen na základě metody faktorové analýzy a regresní analýzy absorbanční matice programem SQUAD(84).

Abstract:

The thermodynamic dissociation constant of hydroxyurea was estimated using potenciometric titration data at 25°C and 35°C. The mixed dissociation constants pKa were estimated on the basis of a goodness-of-fit by the regression analysis of potenciometric data using two programs ESAB and HYPERQUAD. From dependence of dissociation constants on the root of the ionic strenght at 25°C and 37°C by the non-linear regression analysis of the Debye ? Hückel law with the use of the statistic program ADSTAT the thermodynamic dissociation constant pKaT was estimated. The thermodynamic dissociation constants of alendronate sodium and sildenafil citrate was estimated using pH-spectrophotometric titration with UV detection at 25°C and 35°C. By the regression analysis of the absorbance response surface using two different programs SQUAD(84) and SPECFIT32 the mixed dissociation constants pKa were estimated. From dependence of dissociation constants on the root of the ionic strenght at 25°C and 37°C, the thermodynamic dissociation constants pKaT by the non-linear regression analysis of the Debye ? Hückel law in the statistic program ADSTAT were estimated. The chemical model of drugs and the number of light-absorbing species were estimated with use of the factor analysis and regression analysis of absorption matrix using the program SQUAD(84).
