Expression and function of death receptors in human embryonic stem cells and their differentiated derivatives

Abstract:

Receptory smrti poskytují organismu důležitý mechanismus zachování homeostázy, nejen tím, že eliminují pro organismus nebezpečné buňky a chrání ho tak před maligním onemocněním, ale také tím, že se podílí na eliminaci nepotřebných buněk při správném vývoji organismu. Popis exprese a funkce těchto receptorů u lidských embryonálních kmenových (hES) buněk však zatím nebyl uskutečněn. Tato práce podává prvotní popis exprese receptorů smrti a jejich hlavních signalizačních cest u těchto buněk. Zjistil jsem, že u hES buněk, ačkoliv exprimovaly receptory DR4 i DR5 a zároveň neexprimovaly receptory DcR1 a DcR2, nedocházelo po aplikaci ligandu TRAIL k iniciaci apoptózy. Jinak se chovaly neurální progenitorové (NP) buňky a progenitorové buňky neurální lišty (NCP), které rovněž exprimovaly receptory DR4 a DR5 a zároveň neexprimovaly receptory DcR1 a DcR2, u nichž po aplikaci ligandu TRAIL docházelo k masivní apoptóze. Za rezistencí hES buněk k apoptóze indukované ligandem TRAIL zřejmě stojí blokace DISC komplexu proteinem c FLIP. Tato práce otevírá otázku citlivosti hES buněk k apoptóze ve vztahu k receptorům smrti. K detailnímu pochopení této problematiky bude však zapotřebí vést ještě další výzkumy.

Abstract:

Death receptors offer the organism an important mechanism to maintain homeostasis. Not only because they eliminate cells dangerous for the organism, and thus protect it against cancer, but they also participate in eliminating unnecessary cells, during appropriate embryogenesis. However, the expression and function of these receptors in human embryonic stem (hES) cells have not been elucidated, yet. This work is the first to describe the expression of death receptors and their main signalling pathways in these cells. I have found out that in hES cells apoptosis was not initiated after the application of TRAIL, although these cells expressed DR4 and DR5 and yet they did not express DcR1 and DcR2. The neural progenitor (NP) cells and neural crest progenitor (NCP) cells behaved differentely from hES cells. NP and NCP cells also expressed DR4 and DR5 while they did not express DcR1 and DcR2 and there was a massive apoptosis after TRAIL application. The resistance of hES cells to apoptosis induced by TRAIL ligand is probably caused by a blocking of DISC complex by c-FLIP. This work opens a question of sensitivity of hES cells to apoptosis in relation to death receptors. However, to reach a detailed understanding of this topic it will be necessary to do more research.
