Evaluation of the genotoxicity of UV sunscreen

Abstract:

UV filtry používané v kosmetických přípravcích jsou denně aplikovány na kůži zejména v letních měsících pro ochranu před škodlivým UV zářením, proto je důležité se zabývat jejich případnými biologickými efekty a znát jejich mechanismy působení. V řadě studií bylo prokázáno, že některé UV filtry, patřící paradoxně mezi nejpoužívanější aktivní složky opalovacích krémů, působí na endokrinní systém. Mezi tyto UV filtry patří také EHMC (trans EHMC isomer), který na světle podléhá fotodegradaci, při níž vzniká méně stabilní optický isomer cis EHMC. Toxikologické studie se doposud zabývaly jen isomerní formou trans EHMC a informace o fotoproduktu cis EHMC chybí. Tato diplomová práce se zabývá hodnocením rozdílných odpovědí obou isomerů ve vybraných testech toxicity. Jelikož cis EHMC není dostupný jako standard, bylo nutné vyvinout metodiku ozařování EHMC pomocí UV světla a metodu separace cis isomeru z ozářené směsi. Z důvodu ověření přesných koncentrací v průběhu testování byly isomery cis i trans analyzovány pomocí GC FID. Byla vyvinuta metodika separace cis EHMC, kdy bylo dosaženo velmi vysokého výtěžku cis isomeru o čistotě více než 98 %. Cis EHMC byl testován spolu se standardem EHMC na modelech genotoxicity UmuC test a SOS Chromotest. Cílem práce bylo také zavést inovovanou metodu UmuC testu s použitím substrátu CPRG do laboratoře RECETOX. Cis EHMC a standard EHMC byl rovněž testován na estrogenní aktivitu na buňkách MVLN a HeLa9903. V testech estrogenity nebyl estrogenní potenciál EHMC potvrzen. Standard EHMC a izolovaný cis isomer vykázaly rozdílné odpovědi v testech genotoxicity. Je tedy potřeba se této látce věnovat v dalších studiích.

Abstract:

UV filters used in cosmetic products are applied to the skin daily in the summer months to protect against harmful UV rays, so it is important to consider the biological effects and know their mechanisms of action. a number of studies have shown that some UV filters, paradoxically the most widely used sunscreens, have an influence on the endocrine system. Among these UV filters belong also EHMC (trans isomer EHMC) that is subject to photodegradation under UV light and in this process is created less stable optical isomer cis EHMC. Toxicological studies have examined only isomeric form of trans EHMC and information about photoproducts cis EHMC is missing. This thesis deals with the evaluation of different responses of the two isomers in selected toxicity tests. Since cis EHMC is not available as a standard, it was necessary to develop a methodology EHMC irradiation with UV light and the method of separation of cis isomer from the irradiated mixture. In order to verify the exact concentrations during the testing were the isomers cis and trans analyzed by GC FID. It was developed methodology separation of cis EHMC, achieving a very high yield of cis isomer with a purity of more than 98%. Cis EHMC was tested together with the standard model of EHMC on two models of genotoxicity: UmuC test and SOS Chromotest. The aim of this thesis was to introduce to the laboratory RECETOX an innovative method of UmuC test using the substrate CPRG to the. Cis EHMC and standard of EHMC was also tested for estrogenic activity in cells MVLN and HeLa9903. In the tests of estrogenicity wasn’t confirmed the estrogenic potential EHMC. Standard of EHMC and isolated cis isomer showed different responses in tests of genotoxicity. It is therefore necessary to devote to this substance in further studies.
