Determination of selected pharmaceutical compounds by infrared spectroscopy

Abstract:

Tato práce se zabývá možností využití infračervené spektrometrie v blízké oblasti spektra (NIR) při kontrole kvality výroby Paclitaxelu ve firmě Teva Czech Industries spol. s r. o. Vzorky z průběhu chromatografického čištění zahuštěného primárního extraktu jehličí tisu kanadského jsou kontrolovány na obsah taxanů finančně náročnou vysokoúčinnou kapalinovou chromatografií. Také je stanovován obsah sušiny. Byla zkoumána možnost získání těchto dat z měření NIR spektra. V softwaru TQ Analyst byl vytvořen výpočetní model pomocí metody PLS (Partial Least Squares). Kalibrační model obsahuje více než 490 spekter. Bylo zjištěno, že korelační koeficienty a chyby RMSEC a RMSEP pro stanovení taxanů, konkrétně Paclitaxelu a 13-acetyl-9DH-Baccatin III jsou nedostatečné. Model v omezené míře reaguje na obsah sušiny. V tomto případě koeficient chyby RMSEC měl hodnotu 0,9531 a hodnota koeficientu chyby RMSEP byla 0,9231.

Abstract:

This Master thesis deals with examination of the possibility of infrared spectroscopy utilization in a near region of the spectrum during quality control of Paclitaxel manufacturing in Teva Czech Industries spol. s r.o. Samples obtained through chromatographic purification of the primary extraction of the Canadian yew needles are assayed for the amount of taxanes by using financially demanding high performance chromatography. The amount of dry matter is also determined. The possibility of obtaining of these data through the measuring of the NIR spectrum was researched. A computing model was created through the TQ Analyst software using the PLS (Partial Least Squares) method. Calibration model contains more than 490 spectra. The the correlation coefficients and error values RMSEC and RMSEP for the determination of taxanes, specifically Paclitaxel and 13-acetyl-9DH- Baccatin III, is insufficient. Model reacts to the content of dry matter on a limited basis. In this case coefficient of RMSEC had the value 0,9531 and coefficient of RMSEP had the value 0,9231.
