The role of p63 isoforms in tumor cell chemosensitivity

Abstract:

Protein p63 je transkripční faktor patřící do rodiny p53, který se uplatňuje při embryonálním vývoji, tumorigenezi a odolnosti nádorových buněk vůči chemoterapeutickým látkám. Gen TP63 kóduje 12 izoforem tohoto proteinu, které se liší alternativním sestřihem C-konce a přítomností různých transaktivačních (TA) domén. TAp63 izoforma působí jako nádorový supresor a může aktivovat transkripci apoptotických genů podobně jako protein p53. ΔNp63 izoforma s kratší TA doménou bývá označována jako onkogen podporující růst nádorových buněk a její zvýšená hladina byla nalezena u mnoha typů nádorů. Příkladem nádoru se zvýšenou expresí ΔNp63 je triple-negativní karcinom prsu, pro který je charakteristická absence progesteronového a estrogenového receptoru a nízká exprese HER2. Tento typ nádoru je spojován se špatnou prognózou a časným návratem onemocnění. V praktické části této bakalářské práce byl testován vliv exprese ΔNp63 na odolnost nádorových buněk k chemoterapeutikům a na regulaci buněčného cyklu.

Abstract:

p63 protein is a transcriptional factor, that belongs to p53 family, and is involved in embryonic development, tumorigenesis and chemosensitivity of tumor cells. The TP63 gene encodes 12 isoforms of this protein that differ at the C-terminus or in the structure of transactivation (TA) domain. TAp63 isoform acts as a tumor suppressor and could activate some of the proapoptotic genes similar to p53. ΔNp63 with alternative TA domain is usually considered as an oncogene supporting tumor cell proliferation. This isoform was found to be overexpressed in some cancer types, for example in triple-negative breast carcinomas (TNBC). These carcinomas are characterized by the absence of progesterone and estrogen receptors and lack of HER2 overexpression. This breast cancer type is connected with worse prognosis and early relapses. In the experimental part of this bachelor thesis we tested the effect of the overexpression of ΔNp63 on chemoresistance of cancer cells and its influence on cell cycle regulation.
