Molecular characterization of dental and peridental mesenchymal cell populations

Abstract:

Mezenchymální buňky představují dynamickou populaci buněk, která se účastní mnoha významných vývojových procesů, kam patří i odontogeneze a s ní související osteogeneze. V odontogenezi má mezenchym důležitou roli, jelikož jeho interakcí se zubním epitelem je utvářena zubní papila a zubní folikul. Zubní folikul dává v pozdějších stádiích odontogeneze vznik periodontálním strukturám, mezi něž řadíme cementum, alveolární kost a periodontální vazy. Tyto struktury jsou nezbytné pro ukotvení zubu v čelisti. Alveolární kost se utváří intramembranózní osifikací s přímým podílem mezenchymálních buněk zubního základu. Osifikace se obecně účastní široká skupina genů, které se zapojují do diferenciace osteoklastů, osteoblastů a osteocytů. Většina dosavadních poznatků se však týká kostí tvořených endochondrální osifikací. Tato diplomová práce je zaměřena na molekulární screening osteogenního potenciálu mezenchymálních buněk dentálních a peridentálních populací během iniciace tvorby alveolární kosti. Základem analýz bylo relativní srovnání genové exprese s využitím metody PCR Array zahrnující profil 84 genů souvisejících s osteogenezí. Výsledky ukázaly statisticky signifikantní nárůst exprese genů Fgf3, Ctsk, Icam1, Mmp9, Itga3, Tuft1 a celé řady kolagenů (Col1a2, Col3a1, Col7a1, Col12a1, Col14a1) a pokles genů Col2a1, Sox9, Smad2 a Vegfb. Subtilnější změny byly patrné i u dalších genů. Výzkum identifikoval kandidátní geny, které mohou mít zásadní význam pro zahájení tvorby alveolární kosti a je základem následujících funkčních studií.

Abstract:

Mesenchymal cells represent a dynamic cell population of which is involved in many important developmental processes including odontogenesis and related osteogenesis. The mesenchyme plays a key role in odontogenesis due to its interaction with the dental epithelium that forms the dental papilla and dental follicle. In the later stages of odontogenesis the dental follicle is a source of periodontal structures which include the cementum, alveolar bone and periodontal ligaments. These structures are necessary to anchor the tooth in the jaw. The alveolar bone originates by intramembranous ossification with direct participating of the mesenchymal cells of the tooth germ. Ossification is in general accompanied by expression of a large group of genes that are involved in differentiation of osteoclasts, osteoblasts and osteocytes. The most of current findings, however, deal with bones formed by endochondral ossification. This thesis is focused on the molecular detection of osteogenic potential of the mesenchymal dental and peridental populations of cells during intiation of alveolar bone formation. The basis of our analyses was a relative comparison of gene expression using PCR Array including a profile of 84 genes associated with osteogenesis. The results showed a statistically significant increase in the expression of genes Fgf3, Ctsk, Icam1, Mmp9, Itga3, Tuft1 and a wide range of collagens (Col1a2, Col3a1, Col7a1, Col12a1, Col14a1) and a decrease of Col2a1, Sox9, Smad2 and Vegfb. Moreover additional subtle changes were observed for other genes. The research has identified candidate genes that may be crucial for the initiation of the alveolar bone formation and is a background for the following functional studies.
