Developmental toxicity of cyanobacterial metabolites and their mixtures

Abstract:

Sinice mohou produkovat širokou škálu potenciálně škodlivých látek, z nichž bylo mnoho podrobně popsáno. Tato práce se zaměřuje na retinoidy, jakožto jednu ze skupin metabolitů produkovaných některými druhy sinic (např. Microcystis aeruginosa nebo Aphanizomenon klebahnii). Testována byla vývojová toxicita těchto látek na modelovém organismu Danio rerio za použití rybího akutního embryonálního testu (zFET) po dobu 120 hodin. Ve vzorcích odebraných v prostředí se nejčastěji vyskytovalo těchto devět látek: ATRA, 9cis-RA, 13cis-RA, 4OH-ATRA, 4keto-ATRA, 5,6epoxy-ATRA, retinal, 4keto-retinal a retinol. Jelikož se tyto látky ve vodách vyskytují v různých poměrech a je tedy složité přesně otestovat konkrétní složení dané směsi, byla každá z těchto látek otestována samostatně pro určení relativní potence (vůči ATRA). Na závěr byla otestována také rekonstituovaná směs odpovídající složení extraktu biomasy odebraného v nádržích s rozvinutou biomasou sinic v České republice. Díky tomu bylo možné zjistit, zda relativní potence čistých látek korespondují s podílením se jednotlivých látek na vývojových efektech predikovaných na základě dříve popsaných in vitro výsledků a chemické analýzy. Všechny látky kromě retinolu vykazovaly teratogenní účinky již na nízkých koncentracích (v rámci jednotek až desítek µg/L), retinol byl otestován až po 1 g/L, ani na této koncentraci však nepůsobil žádné efekty. Rekonstituovaná směs odpovídala předpokládaným efektům v porovnání s přírodním extraktem a bylo tedy prokázáno, že negativní důsledky na embryonální vývoj obratlovců v tomto vzorku jsou způsobeny retinoidy. Za modelovou látku retinoidů je považována ATRA, jelikož má v organismu obratlovců nezastupitelnou roli a mechanismus jejího působení je z celé skupiny těchto látek nejvíce prozkoumaný – proto byl mimo teratogenity otestován také její vliv na změnu exprese genů signálních drah RAR a RXR pomocí metody qPCR. Testovány byly dvě skupiny embryí, kdy jedna byla exponována vyšší koncentraci ATRA (0,5 µg/L) po dobu 24 hodin a druhá naopak nižší koncentraci (0,22 µg/L) po dobu 120 hodin, ani u jedné skupiny se však neprojevily výrazné změny v genové expresi oproti kontrolním vzorkům. Statisticky významně (na hladině 0,05) vyšlo pouze mírné zvýšení exprese u genu fgf8 na 24 hodinách expozice. V této práci tedy bylo prokázáno, že efekty na embryonální vývoj modelového organismu Danio rerio v rekonstituované směsi připravené dle přírodní biomasy sinic jsou způsobeny retinoidy, a i přes výzvu, kterou obecně testování směsí přináší, je možné poukázat na konkrétní látky, jejichž působením dochází k narušení přirozeného vývoje obratlovců.

Abstract:

Cyanobacteria are known to produce a wide spectrum of potentially harmful compounds, out of which many of them were described in detail in previous studies. In this thesis we focus on retinoids as one group of active metabolites produced by certain species of cyanobacteria (e.g. Microcystis aeruginosa or Aphanizomenon klebahnii). Developmental toxicity of selected set of retinoid compounds was tested by means of 120 hours long fish embryonal toxicity test with Danio rerio (zFET). In the environment there were nine compounds detected most frequently in relation with occurrence of cyanobacteria: ATRA, 9cis-RA, 13cis-RA, 4OH-ATRA, 4keto-ATRA, 5,6epoxy-ATRA, retinal, 4keto-retinal and retinol. These compounds occur in various concentrations and ratios in natural waters. This makes it difficult to test every particular constitution of given mixtures, so each of those nine compounds was tested separately to establish their relative potency (relative to ATRA). Subsequently, a reconstituted retinoid mixture corresponding to the composition of biomass extract from developed cyanobacterial bloom collected from water body in the Czech Republic was tested. This allowed to test whether the relative potencies correspond with contribution of individual compounds to the developmental effects predicted from previously described in vitro results and chemical analysis. All compounds showed teratogenic effects already at low concentrations (units to tens µg/L) except for retinol, which was tested up to 1 g/L, but still did not show any visible effects. The effects of the reconstituted mixture corresponded with the anticipated effects based on the results obtained for the natural extract and in vitro potencies of the retinoids. The results also support that the adverse effects of this sample on the embryonal development of vertebrates were caused by retinoids. ATRA is assumed to be model compound for retinoids, due to its irreplaceable role in vertebrate organisms and its mechanisms of action being the best described among the retinoids. Therefore, ATRA as the fundamental compound was beside the teratogenicity tested for relative changes in gene expression of RAR and RXR signalling pathways by means of qPCR. Two groups of embryos were used – one was exposed shortly (24 hours) to higher concentration of ATRA (0,5 µg/L) and the other one was in reverse exposed to lower concentration for a longer amount of time (0,22 µg/L, 120 hpf), but none of them showed any distinct difference in relative gene expression compared to the control samples. The only statistically significant (significance level 0,05) relative change was slight upregulation of the fgf8 gene at 24 hours of exposition. It was proven in this thesis that the effects of the reconstituted mixture, which was prepared according to the sample of cyanobacterial biomass collected in the environment, on embryonal development of Danio rerio are caused by retinoids and even though the testing of mixtures in general is rather difficult, it is possible to point out particular compounds causing disruption in vertebrates development.
