Induction of epidermal UV-shielding during development of wild-type barley leaves and its mutant chlorina f2

Abstract:

Pro experiment byly použity rostliny ječmene jarního (Hordeum vulgare L.cv.Bonus), a to divoký typ (wild type; WT) a jeho mutant chlorina f2 (clo f2), na kterých byla měřena fluorescence chlorofylu a pomocí fluorimetru Xe-PAM (Heinz Walz, Effeltrich, Německo). Pro vyhodnocení míry UV-stínění u rostlin ječmene jarního bylo použito poměru mezi fluorescencí chlorofylu a excitovanou v modré (B), resp. ultrafialové (UV) oblasti spektra (F-B/F-UV). Cílem této práce bylo stanovení míry epidermálního UV-stínění u výše zmíněných rostlin pěstovaných při dvou ozářenostech (HI, 1000?mol m-2s-1 ; LI, 50?mol m-2s-1 ). Cíle: Porovnat míru UV-stínění divokého typu ječmene jarního (WT) a jeho mutanta chlorina f2 (clo f2), během vývoje a při dvou ozářenostech (HI,LI). Zabývat se gradientem relativní koncentrace UV-stínících látek podél listů. Zároveň se zaměřit na kinetiku těchto změn během vývoje rostlin.

Abstract:

For this experiment , the wild-type spring barley plants (Hordeum vulgare L.cv.Bonus) and its mutant chlorina f2 were used for the measurements of chlorophyll a fluorescence using Xe-PAM fluorimetr. For evaluation of the UV-shielding rate of barley plants, the ratio between fluorescence of chlorophyll a excited in blue (B) and ultraviolet (UV) spectral region (F-B/F-UV) was used. The purpose of this experiment was to determine the rate of epidermal UV-shielding of these plants, which were grown at the two irradiances (HI, 1000?mol m-2s-1 ; LI, 50?mol m-2s-1 ). Aims: Estimation of changes in UV-screening across particular parts of barley leaf and Chl b-less barley mutant chlorina f2 exposed to two levels of irradiance during development. Process result in form of longitudial gradients of UV-screening pigments relative concentration. At the same time deal with kinetics of these changes during development of plants.
