Numerical and structural changes of chromosomes

Abstract:

Chromozomálními a numerickými aberacemi se rozumí mutace projevující se na úrovni struktury jednotlivých chromozomů či změně počtu chromozomů v karyotypu. Takové mutace vznikají spontánně nebo jsou indukovány mutageny prostředí. Předpokladem pro vznik strukturální aberace je dvouřetězcový zlom molekuly DNA. Chromozomální zlom může být restituován nebo reparován prostřednictvím reparačních systémů. V některých případech však proběhne reparace nesprávně a generuje chromozomy s nestandardní strukturou. Numerické aberace jsou zpravidla důsledkem nerovnoměrného rozdělení chromozomů do dceřiných buněk při buněčném dělení. Chromozomální aberace ovlivňují životaschopnost i fitness jejich nositelů. Mutace vedoucí ke ztrátě životně důležitých genů bývají téměř vždy letální. Duplikace genetického materiálu v karyotypu může jedinci poskytnout evoluční výhodu v podobě matrice pro tvorbu nových genů a tím přispět k adaptaci a speciaci druhu. Některé polyploidní organizmy jsou využívány ve šlechtitelství za účelem zvýšení zemědělských výnosů. Tato práce podává celkový přehled poznatků týkajících se problematiky numerických a strukturálních aberací, vypracovaný za použití recentní odborné literatury. Hlavní pozornost je věnována mechanizmům vzniku chromozomálních odchylek, metodám jejich detekce, klasifikace a významu jak pro teoretické poznání, tak pro praktické aplikace.

Abstract:

Chromosomal and numerical aberrations are mutations manifested at the level of the structure of individual chromosomes or changes in the number of chromosomes in the karyotype. Such mutations arise spontaneously or are induced by mutagenic enviroment. A prerequisite for the formation of structural aberration is double strand break of DNA. A chromosomal break can be restored or repaired through repair systems. In some cases, such as incorrect reparation, chromosomal abnormalities can be generated. Numerical aberrations are usually the result of the uneven distribution of chromosomes into daughter cells during cell division. Chromosomal aberrations are affecting the viability and fitness of their carriers. Mutations resulting in a loss of essential genes are almost always lethal. A duplication of genetic material in the karyotype can provide an individual with an evolutionary advantage in the form of a matrix for creation of new genes and thereby lead to the adaptation and speciation of the species. Some polyploid organisms are used in breeding to increase agricultural yields. This thesis provides a general overview of the knowledge related to the issue of the numerical and structural aberrations, using recent literature. The main attention is paid to the mechanisms of formation of chromosomal aberrations, methods of their detection, classification and significance for the both theoretical knowledge and practical applications.
