New methods for evaluating the effect of antibiotics on the growth properties of therapeutic bacteriophages

Abstract:

V 21.století se stále častěji potkáváme s antibiotickou rezistencí bakteriálních kmenů, včetně kmene Staphyloccocus, jejichž infekce jsou obtížně léčitelné a mohou způsobit smrt pacienta. Alternativním přístupem boje s těmito infekcemi je fágová terapie. Ukazuje se, že fágová terapie by mohla najít uplatnění nejen jako samostatné terapeutikum, ale mohla by být také využita v určitých kombinacích s antibiotiky, kdy byl pozorován synergický efekt těchto dvou agens (Phage Antibiotic Synergy, PAS). Tato práce se věnuje metodám vhodným k výzkumu tohoto efektu. V experimentální části byla ověřena citlivost kmenů Staphyloccocus k různým druhům antibiotik a bakteriofágům P68 a 812h1. Synergický efekt byl ověřen jak plotnovými metodami, tak novými spektrofotometrickými metodami a kvantitativní PCR (qPCR). Využity byly také nové metody hodnocení zahrnující Index virulence, Skóre fága a Procento redukce. Tato hodnocení potvrdila synergické působení fosfomycinu na růstové vlastnosti fága P68.

Abstract:

In 21 century, we faced antibiotic-resistant bacterial strains more frequently, including the strain Staphylococcus, which infections are difficult to treat and could cause the death of the patient. An alternative approach how to fight with these infections is phage therapy. It occurs that phage therapy could find an application not just like only therapeutic, but it could be used in specific combinations with antibiotics, in which were observed synergistic effects of both agents (Phage Antibiotic Synergy, PAS). This diploma thesis is dedicated to new methods suitable for evaluating this effect. The experimental part of the thesis evaluated the sensitivity of bacterial strain Staphyloccocus to diverse antibiotics and bacteriophages P68 and 812h1. The synergistic effect was proved by the double-layer agar method and by new methods based on spectrophotometric measurements and qPCR. In the determination of effect was employed new methods of evaluation included the Index of virulence, Phage Score, and Reduction percentage. These methods evaluated the synergistic effect of Fosfomycin on the growth properties of phage P68.
