In vitro growth and cultivation of Photorhabdus sp. and its interaction with an entomopathogenic nematodes Heterorhabditis bacteriophora

Abstract:

Tato práce předkládá ucelený popis bakterií rodu Photorhabdus. Teoretická část se zabývá přehledem taxonomického zařazení, předkládá popis morfologie bakteriálních buněk, metabolických vlastností a představení dvou růstových fází, ve kterých se bakterie objevuje. V práci je popsán symbiotický vztah bakterií a entomopatogenních hlístic Heterorhabditis a také patogenní vztah tohoto symbiotického páru k hmyzu. Cílem je zavést metody práce s bakterií Photorhabdus do laboratoře a současně porovnat růstové a patogenní vlastnosti divoké kultury bakterie Photorhabdus, její mutantní formy neschopné tvorby jednoho z bakteriálních lektinů a další linie izolované přímo ze živých hlístic. Jedná se o studium funkce chybějícího lektinu a jeho možný význam pro patogenitu vůči larvám hmyzu, dále hledání rozdílů v potřebách kultivace, v růstových vlastnostech, tvorbě bioluminiscence a rozlišení růstových fází všech vzorků. Metodická část zahrnuje podmínky pro rozlišení růstových fází bakterií na selektivních agarových mediích dle zbarvení a dle tvorby bioluminiscence. Práce se věnuje měření růstových vlastností bakteri-álních kultur na základě optické denzity a intenzity bioluminiscence. Součástí je stanovení počtu bakteriálních buněk ve vzorku a patogenity bakterií vůči larvám zavíječe voskového (Galleria mellonella), kterým bylo injikováno různé množství těchto buněk a byla sledována jejich mortalita. Byly zjištěny statisticky významné odlišnosti kmene bakterií izolovaného přímo ze živých hlístic ve všech měřených parametrech. Naopak nebyl prokázán žádný statisticky významný rozdíl mezi mutantní a divokou kulturou, a to jak v růstových vlastnostech a tvorbě bioluminiscence, tak v míře patogenity. Výsledky této diplomové práce nepotvrdily zapojení chybějícího lektinu v interakci s hmyzím hostitelem.

Abstract:

This paper presents a comprehensive description of bacteria of the genus Photorhabdus. The theoretical part deals with an overview of taxonomic classification, presents a description of the morphology, metabolic properties, and an introduction to the two growth phases. Here are described the symbiotic relationship of bacteria and entomopathogenic nematodes Heterorhabditis and the pathogenic relationship to insects. The aim of this paper is to introduce methods of working with the bacterium Photorhabdus in the laboratory and at the same time to compare the growth and pathogenic properties of wild culture of Photorhabdus, its mutant form incapable of producing one of the bacterial lectins and other lines isolated directly from live nematodes. This is a study of the function of the missing lectin and its possible significance for pathogenicity to insect larvae, as well as the search for differences in cultivation needs, growth characteristics, bioluminescence formation and growth phase differentiation of all samples. The methodical part includes conditions for distinguishing the growth phases of bacteria on selective agar media according to colour and bioluminescence formation. The work is devoted to measuring the growth properties of bacterial cultures based on optical density and bioluminescence intensity. It includes the determination of the number of bacterial cells in the sample and the pathogenicity of bacteria to the larvae of the greater wax moth (Galleria mellonella). Statistically significant differences of the bacterial strain isolated directly from live nematodes were found in all measured parameters. In contrast, no statistically significant difference was found between mutant and wild culture, both in growth characteristics and bioluminescence formation and in the degree of pathogenicity. The results of this diploma thesis did not confirm the involvement of the missing lectin in the interaction with the insect host.
