Determination of nonstructural carbohydrates content by anthrone method

Abstract:

Cílem práce bylo vypracovat rešerši na téma role nestrukturních sacharidů (NSS) u rostlin a popsat významné spektrofotometrické metody stanovení NSS a škrobu u rostlin. V rešeršní části práce je zmíněna role sacharidů ve fotosyntetickém procesu a popsána syntéza sacharózy a škrobu v postasimilačních procesech. Podstatným oddílem teoretické části je rozdělení rostlinných uhlovodíků na strukturní (SC) a nestrukturní (NSC) a další rozčlenění NSC na cukry, fruktany a škroby. A konečně je zmíněna role sacharidů během abiotického stresu u rostlin a vztah mezi sacharidy a reaktivními formami kyslíku (ROS). Cílem experimentální části bylo zavést metodiku extrakce NSS z rostlinného materiálu a stanovení obsahu rozpustných sacharidů a škrobu s použitím anthronového činidla. Bylo zjištěno, že anthronové činidlo je nutno ponechat minimálně 2 hodiny od jeho přípravy stabilizovat. Dále bylo ověřeno, že anthronové činidlo je stabilní i po 6 dnech, avšak každé sadě vzorků je vhodné přidat minimálně dva kalibrační roztoky pro kontrolu stálosti činida. Výsledkem porovnání několika extrakčních postupu, byla zvolená metoda extrakce pomocí extrakčního činidla methanol: chloroform: voda (MCW) a následná analýza s využitím anthronového činidla. Vytvořená metodika byla aplikována na sazenice ječmene setého (Hordeum vulgare L.), které byly kultivovány při třech různých ozářenostech 100, 200 a 400 ?mol?m-2?s-1 (HI, MI, LI). Byly nalezeny průkazné rozdíly v množství NSC mezi HI a LI variantou. Při vysoké ozářenosti bylo syntetizováno větší množství sacharidů, než při ozářenostech nižších. Při dalším experimentu byl ječmen setý (Hordeum vulgare L.) kultivován při třech různých teplotách 12 °C (LT), 20 °C (NT), 35 °C (HT) a kontrastních ozářenostech 50 ?mol?m-2?s-1 (LI) a 1000 ?mol?m-2?s-1 (HI). I za těchto podmínek rostl obsah NSC s ozářeností, ale pozitivní efekt teploty na obsah NSS byl nalezen pouze u vzorků s vysokou ozářeností.

Abstract:

The aim of this bachelor thesis was evaluate the actual literature related to the topic of non-structural saccharides (NSS) in plants and describe relevant spectrophotometric methods for the determination of NSS and starch in plants. The role of carbohydrates in the process of photosynthesis and the synthesis of sucrose and starch in post-assimilation processes are described as well. Plant carbohydrates are divided into structural (SC) and non-structural (NSC), while NSC are further divided into sugars, fructans and starch. Finally, the role of carbohydrates during abiotic stress in plants and the relationship between carbohydrates and reactive oxygen species (ROS) are mentioned. The experimental part of the thesis deals with introduction of the extraction method of NSS and the determination of the soluble carbohydrates and starch using an anthrone reagent. It was found out that the anthrone reagent is stable for at least 6 days, but it must be allowed to stabilize for at least two hours after preparation. Besides that at least two calibration solutions should be included with each set of samples to check the reagent stability. As a result of a comparisons of several extraction methods, the method of extraction using methanol: chloroform: water (MCW) has been chosen for further NSS and starch analysis. Developed method of extraction and subsequent NSS and starch analysis was applied to seedlings of barley (Hordeum vulgare L.), which were cultivated under three different irradiations 100, 200, 400 ?mol?m-2?s-1. The significant differences in NSS content between HI and LI plants was found out. Under high irradiance, more carbohydrates were synthesized in comparison to plants cultivated under low light. In another experiment, barley (Hordeum vulgare L.) was cultivated under three different temperatures 12 °C (LT), 20 °C (NT), 35 °C (HT) and contrast irradiations 50 ?mol?m-2?s-1 (LI) and 1000 ?mol?m-2?s-1 (HI). It was observed that the NSC content increased with irradiation. Whereas the positive effect of temperature on NSS content was observed only in plants grown at high irradiance.
