Study of metal ion substitutions in binding sites of lectins

Abstract:

Tato práce se zabývá studiem lektinů PA-IIL z Pseudomonas aeruginosa a BC2L-A z Burkholderia cenocepacia. Tyto lektiny patří do lektinové rodiny PA-IIL, pro které je charakteristická přítomnost dvou Ca2+ iontů ve vazebném místě zprostředkovávajících vazbu sacharidu. V rámci diplomové práce byla studována možnost záměny vápenatých iontů ve vazebných místech lektinů PA-IIL a BC2L-A za jiné ionty kovů a následně vliv na vazebné vlastnosti lektinů. V případě možného nahrazení Ca2+ iontů za ionty jiných kovů by bylo zajímavé případné využití pro bioremediaci těžkých kovů. V rámci této práce je popsána produkce a purifikace proteinů, pokusy o nahrazení Ca2+ iontů za jiné ionty kovů a testy vazebných vlastností metodou izotermální titrační kalorimetrie. Byly také připraveny krystaly komplexů proteinů se zaměněnými kovy ve vazebném místě. Výsledky naznačují, že Ca2+ lze odstranit a následně nahradit za jiné ionty kovů.

Abstract:

This work focuses on the study of the lectins PA-IIL from Pseudomonas aeruginosa and BC2L-A from Burkholderia cenocepacia. These lectins belong to the PA-IIL lectin family, which is characterized by the presence of two Ca2+ ions in the binding site that mediate sugar binding. The aim of this thesis was to investigate the possibility of substituting the calcium ions in the binding sites of PA-IIL and BC2L-A lectins with other metal ions, and the subsequent effect on their binding properties. If Ca2+ ions could be replaced with other metal ions, this would be interesting for the bioremediation of heavy metals. The work describes the production and purification of proteins, experiments on the substitution of Ca2+ ions with other metal ions, and tests of the binding properties using isothermal titration calorimetry. Crystals of protein complexes with substituted metals in the binding site were also prepared. The results indicate that Ca2+ ions can be removed and subsequently replaced with other metal ions.
