Antimicrobial effects of slime of snail (Gastropoda)

Abstract:

Plži čeledi Achatinidae a Helicidae produkují celou řadu aktivních látek s antimikrobiálním účinkem. Vzhledem k narůstající rezistenci mikroorganismů vůči antibiotikům dochází k neefektivní léčbě infekčních onemocnění. Aktuální řešení tohoto celosvětového problému by mohli představovat plži ze skupiny Gastropoda produkující sliz obsahující látky s antimikrobiálním účinkem. Cílem této práce bylo stanovení antimikrobiálního účinku látek nacházejících se u plžů ze skupiny Gastropoda. V této práci byl stanoven a srovnán antimikrobiální účinek humorální složky slizu plžů druhu Achatina reticulata a Helix aspersa maxima. Antimikrobiální účinek slizu plžů byl testován vůči mikroorganismům: Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa a Candida albicans. Ke stanovení citlivosti mikroorganismů vůči extrahovaným vzorkům slizu u obou druhů plžů byla použitá disková difúzní metoda na Mueller-Hinton agaru. Velikost inhibičních zón testovaných kmenů bakterií byly odečítaný po 24 hodinové kultivaci při 36,5 °C. V teoretické části práce jsou shrnuty dosavadní poznatky týkající se produkce antimikrobiálních látek nacházejících se ve slizu plžů a jejich stěžejní využití ve farmaceutickém a lékařském průmyslu. V praktické části je popsán postup stanovení antimikrobiálních účinků slizu obou druhu plžů vůči jednotlivým testovaným mikroorganismům. Taktéž je v této části zavedena metoda UV sterilizace eliminující mikrobiotu plžů, aniž by byl potlačen účinek antimikrobiálních látek nacházejících se ve slizu. Antimikrobiální účinek slizu byl u obou vybraných druhů plžů prokázán vůči všem testovaným mikroorganismům. Nejvýraznější antimikrobiální účinek u plže druhu Helix aspersa maxima byl prokázán vůči bakteriálním kmenům Escherichia coli, Klebsiella pneumoniae a Pseudomonas aeruginosa. V případě plže druhu Achatina reticulata byl zaznamenán nejvyšší antimikrobiální účinek u bakterií Staphylococcus aureus. Přístup aplikovaný v této práci by v budoucnu mohl vést k bližší charakterizaci a izolaci jednotlivých látek nacházejících se ve slizu plžů pomocí detekčních metod (HPLC, SDS-PAGE, MS).

Abstract:

The gastropods of the Achatinidae and Helicidae families produce a variety of active substances with antimicrobial activity. Due to the increasing resistance of microorganisms to antibiotics, there is ineffective treatment of infectious diseases. Gastropoda mucus-producing snails containing antimicrobial agents could be the current solution to this global problem. The aim of this study was to determine the antimicrobial effect of substances produced by Gastropoda snails. In this work, the antimicrobial effect of the humoral component of the gastric mucus species Achatina reticulata and Helix aspersa maxima was determined and compared. The antimicrobial effect of gastric mucus was tested against these microorganisms: Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa. A disk diffusion method on Mueller-Hinton agar was used to determine the susceptibility of microorganisms to extracted mucilage samples in both gastropod species. The size of the inhibitory zones of the tested bacterial strains was read after 24 hours of culture at 36.5 ° C. The theoretical part of the thesis summarizes the existing knowledge concerning the production of antimicrobial substances found in snail mucus and their main use in the pharmaceutical and medical industry. The practical part describes the procedure for determination of antimicrobial effects of mucus of both species of snails against individual tested microorganisms. Also, in this section, a method of UV sterilization eliminating snail microbiota is introduced without suppressing the effect of antimicrobial substances. The antimicrobial effect of the mucus in both selected snails species was demonstrated against all microorganisms tested. The most pronounced antimicrobial effect in the snail species Helix aspersa maxima was demonstrated against the bacterial strains Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa. In the case of snail species Achatina reticulata, the highest antimicrobial effect was observed in Staphylococcus aureus. The approach applied in this work could in future lead to closer characterization and isolation of individual substances found in the snail mucus by means of detection methods (HPLC, ELFO, MS).
