Hematopoietic bone marrow stem cells: role of cytokines in cultivation and differentiation of hematopoietic stem cells in vitro and their impact in experimental oncology

Abstract:

Cytokines are humoral factors which secure communication within the immunity system and they actively cooperate with other regulation systems of the organism on both specific and non-specific levels at the same time. Today, the term „cytokines” also includes other regulatory proteins, particularly monokines, interleukins, interferons, and growth factors, which can potentially be produced by every nuclear cell while their pleiotropic effects on various cells are maintained. They regulate the range of the costimulation and accessory molecules expression on the cell surface, including the direct influence of homeostasis as well. In hematological malignancies, bone marrow and lymphatic organs are primarily influenced and affected – a generation of immature blood elements occurs there (some of them produce cytokines themselves). Concentration of soluble factors in body fluids can reflect the extent of the charge of an organism by a malignant process. There are also distinct changes in humoral factors concentrations in auto- and alloimmunity diseases. Cytokine concentrations can be used as markers in some diseases. Many experimental studies confirm that the cytokine profile of a patient is connected with his further prognosis. Different therapeutic interferences influence directly or non-directly the immunity mechanisms. This work comprise a brief overview about extensive knowledge about cytokines, their importance, classification and improvement in clinical and experimental therapeutic trials in the field of hematologic and non-hematologic diseases. The author also presents results of her own experimental work concerning cytokines and hematopoietic stem cells.

Abstract:

Cytokíny sú humorálne regulačné faktory, pomocou ktorých medzi sebou komunikujú zložky imunitného systému a tiež ostatné systémy zodpovedné za homeostázu. V súčasnosti sa pojmom cytokíny označujú aj regulačné proteíny typu monokínov, interleukíny, interferóny a rastové faktory, ktoré môžu potenciálne produkovať všetky jadrové bunky. Cytokíny tiež regulujú expresiu kostimulačných a akcesorných molekúl na povrchu buniek. U hematologických malígnych ochorení sú primárne postihnutými orgánmi kostná dreň, resp. lymfatické orgány, kde dochádza k vyplavovaniu nezrelých krvných elementov. Niektoré z týchto nezrelých bunkových štádií sú tiež schopné cytokíny samé produkovať. Koncentrácia solubilných faktorov v telesných tekutinách môže do istej miery reflektovať rozsah postihnutia organizmu nádorovým procesom. Zreteľné zmeny na úrovni koncentrácie humorálnych faktorov prebiehajú aj pri auto- a aloimunitných systémových ochoreniach. Mnohé experimentálne štúdie preukázali význam stanovenia koncentrácie jednotlivých cytokínov u pacientov vo vzťahu k ďalšej prognóze ich ochorenia, ako aj význam rozdielnych terapeutických zásahov priamo či nepriamo ovplyvňujúcich imunitné mechanizmy. Predkladaná práca predstavuje stručný prehľad inak širokej a komplexnej problematiky týkajúcej sa cytokínov, ich významu, klasifikácie, ako aj ich využitia v klinických a experimentálnych štúdiách v oblasti hematologických i nehematologických ochorení. Autor tiež v práci prezentuje vlastné experimentálne výsledky týkajúce sa cytokínov a hematopoetických kmeňových buniek.
