Structure and function of plasmids isolated from methicillin resistant strains of Staphylococcus aureus

Abstract:

Staphylococcus aureus je oportunitní patogen a hlavní příčina mnoha nemocničních a komunitních infekcí. Diplomová práce pojednává o charakterizaci plazmidů u meticilin rezistentních kmenů. Transdukce byla využita k přenosu plazmidů do bezplazmidových laboratorních kmenů. Jako transdukující fágy byly použity fágy sérologické skupiny B. Plazmidy byly popsány molekulárními metodami. Byl potvrzen přenos penicilinázového plazmidu. Přibližná velikost plazmidů byla určena restrikční analýzou a fenotyp byl stanoven pomocí antibiotikové diskové metody.

Abstract:

Staphylococcus aureus is an opportunistic pathogen and the major causative agent of numerous hospital and community acquired infection. The diploma thesis deal with the characterization of plasmids of the staphylococcal methicillin resistant strains. The transduction was used to plasmid transfer in plasmid less recipient strains. As transducing phages were used the phages of the serologic group B. The plasmids were described by molecular techniques. The transfer of pecillinase plasmids was manifested. The approximate size of the plasmids were deternined throught restriction anylysis and the antibiotics resistant phenotype was determined using the antibiotics disc method.
