Bioinformatické vyhledávání GT-C glykosyltransferas v genomu bakterie <I>Mycobacterium tuberculosis</I>

Bioinformatic searching for GT-C glycosyltransferases in <I>Mycobacterium tuberculosis</I>

Abstract:

Tato bakalářská práce se věnuje bioinformatickému vyhledávání genů kódujících hypotetické a necharakterizované proteiny v genomu bakteriálního kmene <i>Mycobacterium tuberculosis</i> CDC1551, které by mohly být dosud neobjevenými glykosyltransferasami s foldem GT-C. Glykosyltransferasy se spolu s dalšími enzymy podílejí na vzniku výjimečně odolné buněčné stěny bakterií <i>M. tuberculosis</i>, která brání léčbě pomocí antibiotik, což může vést např. ke vzniku rezistentních kmenů. Proto je vyvíjena snaha cílit nové léky proti tuberkulóze mimo jiné také na glykosyltransferasy. K jejich vyhledání byl použit bioinformatický nástroj BackPhyre, který slouží k prohledání vybraného genomu podle modelu proteinu, jehož struktura je již vyřešena. Nalezené sekvence byly protříděny na základě vyhledávání v databázích a analýzy programem BLAST. Nakonec byly vytipované proteiny charakterizovány pomocí dalších bioinformatických nástrojů, jako je predikce transmembránových oblastí, predikce 3D struktury, predikce vazebných míst a prohledávání operonů.

Abstract:

This thesis deals with the bioinformatic search for genes that encode hypothetical and uncharacterized proteins in the genome of the <i>Mycobacterium tuberculosis</i> strain CDC1551. These proteins might be undiscovered GT-C glycosyltransferases that, together with other enzymes, are involved in the biosynthesis of an exceptionally resistant cell wall of <i>M. tuberculosis</i>;. This cell wall prevents treatment with antibiotics, which can lead to antibiotic resistance. Therefore, efforts are being made to target new anti-tuberculosis drugs against glycosyltransferases. To find them, we used BackPhyre, a bioinformatics tool for searching the selected genome based on a protein model with a resolved structure. The sequences we found were sorted by results of database search and BLAST analysis. Finally, the selected proteins were characterized using other bioinformatic methods, such as transmembrane regions prediction, 3D structure prediction, ligand binding sites prediction and operon browsing.
