NMR studies of peptides binding rare metal pollutants

Abstract:

Odpad z eletronických zařízení je významným problémem v současném světě. Špatné zacházení s elektroodpadem a jeho zvyšující se produkce představují hrozbu pro životní prostředí a lidské zdraví. Proto je jedním z cílů vědy a technologií vývoj způsobů zpracování elektroodpadu, které by snížilo vliv na životní prostředí a umožnilo získat znovu zužitkovatelné části. Kovy, které jsou vysoce žádané součásti elektronických zařízení, jsou získávány především z primárních zdrojů a jejich recyklace je velmi nízká. Biosorpce je jednou z možností jak získat kovy z elektroodpadu. Je to jeden z hlavních zájmů v Helmholtz Institute Freiberg for Resource Technology v Drážďanech, kde popsali peptidy vázající ionty gallia z roztoku, které by mohly být použity k získání gallia z kontaminovaných vod. V této práci byla zkoumána struktura peptidu SDM-M3 (HTCHIQSCDHLA) ve formách s galliem i bez gallia užitím nukleární magnetické rezonance. Pomocí 2D experimentů 1H-1H NOESY, 1H-1H TOCSY and 1H-13C HSQC byly přiřazeny rezonanční frekvence. Pro charakterizaci struktury byl použit výpočet tendence tvořit sekundární struktury (Secondary structure propensity) a interakční konstanty 3JHN-HA. Získané hodnoty chemických posunů nevykazují výrazné změny mezi formami kromě chemického posunu atomu Cβ residua D9. Secondary structure propensity skóre souhlasí s tvrzením, že konformace peptidu není ovlivněna vazbou gallitých iontů. 13C hodnoty chemických posunů vypovídající o disulfidickém můstku mezi residui C3 a C8, charakteristické trendy 3JHN-HA interakčních konstant a 1H-1H NOE naznačují přítomnost ohnuté konformace páteře peptidu, která by mohla být důležitá pro vazbu gallitých iontů.

Abstract:

The waste from electronic devices is a significant problem in the current world. The poor management and increasing production of the e-waste poses a threat to the environment and human health. Therefore one of the aims of science and technology is the development of a treatment that would minimise the impact on the environment and retrieve reusable parts. Metals, which are highly demanded components of many common devices, are currently obtained mainly from their primary finite sources and their recycling rate is low. One of the possible ways of recovering the metals from e-waste is biosorption. It is one of the main interests at Helmholtz Institute Freiberg for Resource Technology in Dresden, where they identified peptides that bind gallium ions in solution and thus might be used for extracting gallium from e-waste waters. In this work, the structural features of the SDM-M3 (HTCHIQSCDHLA) gallium-binding peptide (with and without gallium) were investigated using nuclear magnetic resonance. Using 2D 1H-1H NOESY, 1H-1H TOCSY and 1H-13C HSQC experiments, the assignment of resonances of the peptide was completed. Secondary structure propensity calculation and 3JHN-HA coupling values were used to characterize its structural features. The obtained chemical shift values do not show significant differences between the two forms except for Cβ chemical shift values of residue D9. Secondary structure propensity scores agree with the finding that the conformation of the peptide is not vastly affected by the binding of gallium ions. 13C chemical shifts values indicating the formation of a disulfide bridge between residues C3 and C8, characteristic trends in JHN-HA coupling values and 1H-1H NOEs indicate that the peptide's backbone adopts a bent conformation, presumably important for the gallium binding.
