Modulation of carboxypeptidase B1 using targeted inhibitors

Abstract:

Karboxypeptidáza B1 (CPB1) je specifický tkáňový protein známý jako marker akutní pankreatitidy, avšak zvýšené hladiny CPB1 byly detekovány i u nádoru prsu. V předchozí kombinované proteomické a transkriptomické analýze byla nalezena zvýšená hladina CPB1 u luminálních A nádorů prsu metastazujících do lymfatických uzlin proti nemetastazujícím. V práci byl studován vliv inhibice CPB1 na migrační a invazivní vlastnosti nádorových buněk MCF-7 overexprimujících CPB1 (MCF7-CPB1). Pro inhibici CPB1 byl použit bramborový karboxypeptidázový inhibitor (PCI), jehož vliv byl studován ve “scratch“ migračních a Transwell migrační a invazivní experimenty. Výsledky ukázaly, že inhibice CPB1 významně snižuje migraci buněk overexprimujících CPB1, stejný trend bych patrný i u invazivních testů. Tentýž inhibitor byl aplikován v analýze ovlivnění protein-proteinových interakcí CPB1 pomocí pull-down assay a kombinace kapalinové chromatografie a hmotnostní spektometrie (LC-MS/MS). Byl identifikován protein kódovaný genem DLST signifikantně downregulovaný pomocí PCI in MCF7-CPB1 cells. Práce ukazuje, že inhibice CPB1 ovlivňuje migraci nádorových buněk produkujících CPB1 a podporuje hypotézu, že protein CPB1 je zapojen do procesu metastazování u nádorů prsu.

Abstract:

Carboxypeptidase B1 (CPB1) is a specific tissue protein known as a marker of acute pancreatitis. However, its increased levels of CPB1 were also detected in breast cancer. In the previous proteomics and transcriptomics analysis, CPB1 was highly expressed in lymph node positive (vs. negative) low grade (G1) breast primary tumors of luminal A subtype. The effect of CPB1 inhibition on migration and invasiveness of MCF-7 breast cancer cell line overexpressing CPB1 (MCF7-CPB1) was studied in this thesis. Potato carboxypeptidase inhibitor (PCI) was used for CPB1 inhibition in scratch migration and Transwell migration and invasive experiments. The data showed that CPB1 inhibition of CPB1 significantly decreases migration and invasiveness of MCF7-CPB1 cells, the same trend was visible in invasive tests. The same inhibitor was applied in the analysis of CPB1 protein-protein interactions modulation using pull-down assay and liquid chromatography-mass spectrometry (LC-MS/MS). A protein encoded by DLST gene was found significantly downregulated by PCI in MCF7-CPB1 cells. This thesis shows that CPB1 inhibition affects migration of cancer cells producing CPB1 and supports the hypothesis that CPB1 is involved in breast cancer metastasis.
