The effects of age and infection pressure on the content of phenolics in tomato

Abstract:

Fenolické látky mají antioxidační, protizánětlivé a protikarcinogenní vlastnosti a mohou být používány jako potenciální léčiva nebo jako prevence kardiovaskulárních a neurodegenerativních chorob nebo také rakoviny. Rostlina se brání před škodlivým UV zářením, parazity, patogeny a predátory nárůstem koncentrací těchto látek. Množství a složení fenolických látek u rajčete ovlivňuje také stáří a infekční tlak. Byla zjišťována celková koncentrace polyfenolů pomocí Folin–Ciocalteuho metody u zdravých listů a u listů napadených bakterií Pseudomonas syringae. Jednotlivé fenolické látky byly stanovovány pomocí metody HPLC. Konkrétně byly sledovány kyselina gallová, kyselina chlorogenová, rutin jakožto hlavní fenolické látky a obranné látky rajčete a fenylalanin jakožto prekurzor sekundárních metabolitů. Kyselina gallová ani kyselina chlorogenová ve vzorcích nebyly nalezeny. Napadené listy vykazovaly obecně vyšší obsah fenolických látek než listy nenapadené. Významný rozdíl mezi napadenými a nenapadenými listy tvořila celková koncentrace polyfenolů a obsah rutinu u mladých rostlin. Významný byl také rozdíl v celkové koncentraci polyfenolů mezi starými a mladými napadenými listy staré rostliny. Stáří mělo také vliv na obsah fenylalaninu u napadených listů, kdy se se zvyšujícím se stářím listu obsah fenylalaninu snižoval. Na obsah rutinu mělo však vliv pouze stáří rostliny, kdy došlo opět ke snížení. U staré rostliny tedy nemělo vliv stáří listů na obsah rutinu.

Abstract:

Phenolic compounds have antioxidant, antiinflammatory and anticarcinogenic properties and could be used as potential pharmaceuticals or as prevention of cardiovascular and neurodegenerative diseases or also cancer. Plant protects itself from damaging UV radiation, parasites, pathogens and herbivores by increasing concentrations of these compounds. For this reason, we had to account the effect of age and infection pressure on phenolic content in tomato. The total polyphenol content was determined by Folin-Ciocalteu method in healthy leaves and in leaves infected by Pseudomonas syringae. Individual phenolic compouds were determined by HPLC method. In particular, gallic acid, chlorogenic acid and rutin were determined as major phenolic compounds and phenylalanine was detected as a precursor of secondary metabolites. Neither gallic acid nor chlorogenic acid were found in the samples. Infected leaves generally contained higher phenolic content than healthy leaves. A significant difference between the infected and healthy leaves was in the total concentration of polyphenols and the content of rutin in young plants. A significant difference was also in the total concentration of polyphenols between the old and young infected leaves of the old plant. Age affected phenylalanine content of the infected leaves, the phenylalanine content decreased with increasing leaf age. However, the rutin content was only influenced by the age of the plant, which again decreased with increasing age. Thus, in the old plant, the age of the leaf did not affect the rutin content.
