Molecular characterization of chosen coagulase negative staphylococci and their clinical significance

Abstract:

Coagulase-negative staphylococci represent extensive heterogeneous group within the genus Staphylococcus. Their importance as causative agents of nosocomial infections and as the reservoir of antibiotic resistance genes is constantly increasing, mainly due to the advances in medicine, the use of indwelling devices and the overuse of antibiotics. Most studies on the pathogenicity of staphylococci have been focused on Staphylococcus aureus, and only a few of the most clinically important species in the coagulase-negative staphylococci group have been well studied. The dissertation thesis addresses the molecular characteristics and typing of selected species of staphylococci and their pathogenic potential to cause disease in humans and animals. The thesis summarizes basic knowledge about the genus Staphylococcus and closely related genus Macrococcus, their occurrence, genome organization, pathogenicity and diagnostic options, based on genomic analyses. The dissertation thesis contributed to the increase of knowledge of the already described staphylococcal species, such as Staphylococcus petrasii and members of the group Staphylococcus intermedius, and to the description and characterization of new, previously unknown species such as Staphylococcus edaphicus and members of the genus Macrococcus. The thesis also includes a description of new mobile genetic elements, including a new genus of bacteriophage, Fibralongavirus, and a description of new phages isolated from Staphylococcus sciuri strains. The possibilities of identification and diagnostics of particular species, the structure of staphylococcal genomes and the presence of mobile genetic elements, which significantly contribute to the evolution, adaptation and pathogenicity of members of the genus Staphylococcus, are discussed. The dissertation thesis is executed in the form of an annotated collection of individual publications.

Abstract:

Koaguláza negativní stafylokoky představují velmi širokou heterogenní skupinu v rámci rodu Staphylococcus. Jejich význam jako původců nozokomiálních infekcí a rezervoáru genů rezistence k antibiotikům neustále narůstá, především díky pokroku v medicíně, zavedeným tělesným náhradám a nadužívání antibiotik. Většina studií zaměřena na patogenitu stafylokoků se věnuje Staphylococcus aureus a v rámci skupiny koaguláza negativních stafylokoků je dobře prostudovaných pouze pár klinicky nejvýznamnějších druhů. Dizertační práce se věnuje molekulární charakteristice a typizaci vybraných druhů stafylokoků a jejich patogennímu potenciálu způsobit onemocnění člověka a zvířat na základě genomických analýz. Práce shrnuje základní poznatky o rodě Staphylococcus a jejich blízko příbuzném rodě Macrococcus, jejich výskytu, organizaci genomu, patogenitě a možnostech jejich diagnostiky. Dizertační práce přispěla k rozšíření znalostí o již popsaných stafylokokových druzích, jako jsou Staphylococcus petrasii a členové skupiny Staphylococcus intermedius a k popisu a charakterizaci nových, dosud neznámých druhů jako Staphylococcus edaphicus a zástupců rodu Macrococcus. Součástí práce je i popis nových mobilních genetických elementů včetně nového rodu bakteriofága Fibralongavirus a popis nových fágů izolovaných z kmenů Staphylococcus sciuri. Diskutovány jsou možnosti identifikace a diagnostiky daných druhů, struktura stafylokokových genomů a přítomnost mobilních genetických elementů, které výrazně přispívají k evoluci, adaptaci a patogenitě členů rodu Staphylococcus. Dizertační práce je zpracována ve formě komentovaného souboru jednotlivých publikací.
