Binding properties of p53 protein mutants and their regulation

Abstract:

V této bakalářské práci se věnujeme schopnosti vazby proteinu p53 a jeho mutantů na cílové sekvence a superhelikální DNA. Vazba na DNA je kritická pro vyvolání exprese různých genů jako odpovědi na její poškození. Teoretická část této práce vysvětluje úlohu proteinu v regulaci buněčného cyklu a při apoptóze, popisuje různé oblasti proteinu, jakým způsobem se zapojují do vazby s DNA a také mutace proteinu a jejich důsledky. V experimentální části je pak prokázána schopnost vazby na DNA i přes to, že doména zodpovědná za vazbu na specifickou oblast DNA je poškozena.

Abstract:

In this thesis we study binding ability of protein p53 and its mutants in the target sequence and superhelical DNA. Binding to DNA is crucial for induction of expression of various genes in response to DNA damage. The theoretical part of this thesis explains the role of protein in cell cycle control and apoptosis, describes different sequences of protein and how they are involved in DNA binding and also the protein mutations and their consequences. In the experimental part is then demonstrated the ability of DNA binding, despite the fact that domain responsible for binding to specific region of DNA is damaged.
