Spruce and pine needles as passive samplers and tools for assessing the genotoxic effect of atmospheric contaminants

Abstract:

V průmyslově vyspělých zemích je do životního prostředí nepřetržitě uvolňováno velké množství chemických látek. Ty jsou poté distribuovány v různých složkách životního prostředí (vzduch, voda, půda a biota), kde mohou být degradovány, přeměněny nebo uloženy. Jedna skupina těchto chemických látek jsou perzistentní organické látky (POPs). Jsou nebezpečné pro živé organismy kvůli jejich karcinogenním, mutagenním, teratogenním, neurotoxickým či imunosupresivním vlastnostem. Tyto sloučeniny mají obecně tendenci setrvávat v prostředí po dlouhou dobu a podléhají dálkovému transportu, mohou být akumulovány v lipidových tkáních se schopností biomagnifikace v potravních řetězcích všech trofických úrovních. POPs v atmosféře jsou běžně monitorovány s využitím technik aktivního a pasivního vzorkování ovzduší. Další možnou metodou pro vzorkování ovzduší je použití rostlin charakteristických pro danou vzorkovanou oblast. Biomonitoring kvality ovzduší s použitím vegetace je efektivním, levnějším a lépe dostupným nástrojem pro odhad úrovně znečištění ovzduší. Pro stanovení úrovně znečištění ovzduší POPs byli nejčastěji využívány jehlice nahosemenných rostlin, lišejníky a mechy. Jehličnany jsou široce rozšířené a stále zelené rostliny a mohou tedy kontinuálně akumulovat POPs v řádech roků a jsou tedy ideálním nástrojem pro dlouhodobé sledování trendů POPs na rozlehlé území nebo na vzdálených a špatně přístupných místech. Řada toxických a zdraví škodlivých látek se vyskytuje v životním prostředí ve velmi nízkých koncentracích a jejich identifikace chemickou analýzou může být obtížná a v některých případech není vůbec možná. Použití biologických testů k identifikaci potenciálních rizik pro živé organismy je jedním ze způsobů, jak získat informace i o výskytu POPs ve velmi nízkých hladinách. Informace o kontaminaci životního prostředí (chemická analýza) a účincích na živé organismy (biologický test) mohou tedy poskytnout celkový obraz o znečištění ovzduší, možných zdrojích a rizicích pro lidské zdraví. Teoretická část této disertační práce obsahuje stručný popis jehlic nahosemenných rostlin jakožto pasivního vzorkovače ovzduší. Dále se teoretická část věnuje stručnému přehledu biomonitoringu genotoxických látek a popis nejčastěji používaných testů genotoxicity. Praktická část této disertační práce zahrnuje celkem pět studií. První publikace zahrnuje srovnávací studii zaměřenou na účinnost ukládání POPs v borovicových jehlicích, aktivních a pasivních vzorkovačích (Studie č. 1). Následující Studie č. 2 se zaměřuje na vývoj metodiky odběru vzorků a analýzy borovicových jehlic. Předmětem Studie č. 3 jsou časové trendy vybraných skupin POPs v jehlicích borovice černé (Pinus nigra) a stanovení možných nejistot srovnáním s odpovídajícími vzorky pocházející z aktivního a pasivního vzorkovače ovzduší s použitím polyuretanové pěny (PUF). Distribuce POPs ve vysokohorských oblastech a korelace mezi nadmořskou výškou, koncentrací POPs a genotoxickým zatížením daných lokalit byla cílem studie č. 4. Ve studii č. 5 byly prezentovány dva druhy jehličnanů - smrk a borovice jakožto pasivní vzorkovače ovzduší a jejich možné využití při hodnocení genotoxického efektu znečišťujících látek obsažených v takto odebraných vzorcích ovzduší. Tento doktorský výzkum jednoznačně prokázal, že jehlice nahosemenných rostlin jsou vhodným nástrojem pro hodnocení kvality ovzduší i pro stanovení genotoxického potenciálu znečištění ovzduší.

Abstract:

In industrialized regions, large concentrations of chemicals are emitted continuously to the atmosphere. Once released, these chemicals are distributed over different environmental compartments (air, water, soil, and biota), where they can be degraded, transformed or accumulated. One group of these chemicals are persistent organic pollutants (POPs). To be listed as a POP, a compound must be bioaccumulative, capable of LRT, persistent and have toxic potential. These substances are of both natural and anthropogenic origin. Most of these compounds are hazardous to living organisms due to their carcinogenic, mutagenic, teratogenic, neurotoxic and immunosuppressive effects. In general, these compounds have very low aqueous solubility, high n-octanol-water partition coefficients, and low vapor pressure and tend to bioaccumulate in lipid tissues. Furthermore, they are biomagnified within food-chains. POPs in the atmosphere are conventionally monitored by active and passive air sampling techniques. Vegetation has also demonstrated the ability to accumulate POPs. The monitoring of vegetation is an effective, cheap and easily available tool for estimating atmospheric contamination levels. The spatial distribution of POPs has been most often investigated in coniferous needles, lichens, and mosses. Conifers are widespread and evergreen; they can accumulate POPs for several years and can be used for long-term monitoring over a broad spatial area or at remote and poorly accessible locations. Biomonitoring also allows short term retrospective analysis as needles remain on trees up to 4-5 years. Many toxic and harmful substances and their adverse health effects may be difficult to identify or assess on the basis of chemical analysis alone, partly due to their low concentrations in the environment. The use of bioassays to identify potential risks to living organisms is one of the ways to complete this information. Therefore, combined information about environmental contamination (chemical analysis) and effects on living organisms (bioassay) can give an overall picture about air pollution, including its possible sources and human health risks. The theoretical part of this thesis contains a brief description of the use of coniferous needles as passive air samplers followed by a description of the conventionally used bioassays dealt with by the presented studies. The practical part contains five different studies. Study n.1 compares the efficiency of POPs sequestration in pine needles and in active and passive samplers. Study n.2 focuses on the development of the methodology for the sampling and analysis of pine needles. Study n.3 concerns the temporal trends of selected POPs groups in pine needles (Pinus nigra) and the critical judgement of uncertainties by comparing pine needles with an active air sampler and a polyurethane foam passive air sampler (PUF-PAS). The spatial distribution of POPs in high mountains and the correlation between altitude, the concentration of POPs, and the environmental genotoxic load was the aim of Study n.4. In Study n.5, two coniferous species – spruce and pine – were used as passive air samplers and their possible use in the evaluation of the genotoxic effect of air pollution was presented. This doctoral research clearly shows that coniferous needles are a feasible tool for assessing atmospheric contamination and for evaluating the genotoxic effect of air pollution.
