Significance of p53/47 protein in tumours

Abstract:

Jednou z izoforem nádorového supresoru p53 je N-koncově deletovaná izoforma označována jako p53/47, ΔNp53 nebo také p47, která postrádá prvních 40 aminokyselin, což se také odráží v odlišných vlastnostech tohoto proteinu. Doposud jsou známy dva mechanismy, které vedou k expresi p53/47 - alternativní sestřih a IRES regulovaná iniciace translace mRNA TP53. Z provedených výzkumů je dnes již zřejmé, že tato izoforma má vliv na regulaci transkripční aktivity p53, ale také se může samostatně uplatňovat při navození zástavy buněčného cyklu v G2 fázi nebo při spuštění apoptotické dráhy. Znalosti o regulaci, aktivitě a signálních drahách, v nichž se protein p53/47 uplatňuje, mohou hrát důležitou roli při objasnění mechanismů, kterými se buňka vyrovnává se stresovými podmínkami a které ovlivňují procesy, jako jsou např. karcinogeneze nebo apoptóza. Při analýze hladiny proteinů p53 a p53/47 v karcinomech mléčné žlázy u skupiny 44 pacientek byla detekována současná exprese obou těchto proteinů v 9 případech, pouze exprese proteinu p53/47 u 1 pacientky. Protein p53 se podařilo stanovit u 30 pacientek. V experimentu byla také prokázána fyziologická relevance modelu kultivace ex vivo. Kultivace nepřesahující 6 hodin plně odrážela p53 signalizaci ve skutečné nádorové buňce. Změny v intenzitě exprese jak p53 tak i p53/47 se po aplikaci uvedených cytostatik lišily, což pravděpodobně úzce souvisí se statutem genu TP53. Lze uzavřít, že regulace exprese p53/47 představuje poměrně komplexní proces, na který mají vliv různé faktory a pro pochopení jeho podstaty bude důležité všechny tyto faktory určit a zjistit jejich účinky a vzájemný vliv.

Abstract:

One of the isoforms of the tumor suppressor p53 is N-terminal deleted isoform, also called p53/47, ΔNp53 or p47 which lacks the first 40 aminoacids. This results in different properties of this protein. So far there are two known mechanisms leading to expression of p53/47 - alternative splicing and IRES-mediated translation of the TP53 mRNA. It is obvious from research that was already done that this isoform influences regulation of the p53 transcriptional activity, however it can also act individually in initiating of the cell cycle arrest in the G2 phase or in initiating of apoptotic pathway. Knowledge of regulation, activity and signal pathways in which the p53/47 protein is active may play an important role in explaining the mechanisms by which the cell respond to stress conditions and which influence processes such as carcinogenesis or apoptosis. The analysis of p53 and p53/47 proteins in mammary gland carcinomas in a group of 44 patients, revealed a simultaneous expression of both these proteins in 9 cases and the expression of only the p53/47 protein in 1 case. The p53 protein was determined in 30 cases. Importantly, a physiological relevance of the ex vivo cultivation model was also proven in the experiment. Cultivation that didn't exceed 6 hours was fully reflecting the p53 signalization in a real tumor cell. Changes in intensity of the expression of both the p53 and the p53/47 were different after application of the aforementioned cytostatics. This fact is probably linked to the status of the TP53 gene. In conclusion, we can say that the regulation of the p53/47 expression represents a relatively complex process that is influenced by various factors and in order to understand its basis, it will be important to determine all these factors and find out about their effects and mutual influence.
