Uncovering genes whose depletion exhibits synthetic lethality with inhibition of haspin kinase

Abstract:

Nízkomolekulární inhibitory kináz mají důležité uplatnění a potenciál jako léčiva. Zároveň patří mezi důležité nástroje v oblasti základního výzkumu, kde mohou vést k porozumění funkce proteinu a jeho validaci jako terapeutického cíle. Zejména v této oblasti je podstatná jejich vysoká selektivita a potence, která je důležitým předpokladem pro správnou intepretaci získaných výsledků. První část této práce se zaměřuje na charakterizaci nově syntetizovaných inhibitorů Haspinu, kinázy, která se podílí na kohezi a segregaci sesterských chromatid během mitózy. Bylo využito několika metod pro otestování efektu inhibitorů na chování buněk. U nových látek se prokázala selektivní inhibice Haspinu s minimální nespecifickou inhibicí a nízkou cytotoxicitou. Druhá část práce navazuje na celogenomový CRISPR/Cas9 screening, který měl za cíl identifikovat geny, jejichž delece spolu s inhibicí Haspinu vede k syntetické letalitě. V této práci byly pomocí analýzy obohacení signálních drah (angl. pathway enrichment analysis) popsány nejvýznamnější geny s tímto potenciálem. Dále byla syntetická letalita vybraných genů experimentálně validována kombinací RNA interference s inhibicí Haspinu.

Abstract:

Small-molecule inhibitors play an important role in both translational medicine as well as in basic research. Especially for the latter, the selectivity and potency of the inhibitors is crucial for elimination of the effects of non-specific inhibition and correct data interpretation. This work focuses on the characterisation of newly synthesised inhibitors of Haspin, a kinase involved in the regulation of sister chromatid cohesion and segregation during mitosis. Several cell-based assays were employed to elucidate various effects of the inhibitors on cell behaviour. They proved to selectively inhibit Haspin with minimal off-target effect and cytotoxicity. The second half of this work is a follow-up of a genome-wide CRISPR/Cas9 knock-out screen aimed at discovering synthetic lethal interactions with Haspin inhibition. The description of the highest-ranking genes was performed with pathway enrichment analysis. Furthermore, a combined treatment of RNA interference and Haspin inhibition was employed to validate the potential synthetic lethal interaction experimentally.
