Detection of Mycoplasma hominis and Ureaplasma urealyticum in the urogenital tract of infertile couples by molecular-biology methods

Abstract:

Cílem práce je stanovení frekvence výskytu Mycoplasma hominis a Ureaplasma urealyticum v urogenitálním traktu mužů a žen, léčených v Centru asistované reprodukce SANUS Pardubice. Uvedené mikroorganismy byly prokazovány druhově specifickou polymerázovou řetězovou reakcí. Současně byl sledován vliv kontaminace Mycoplasma hominis a Ureaplasma urealyticum na kvalitu ejakulátu. Výsledky byly následně porovnány s obdobně zaměřenými studiemi. Celkově bylo vyšetřeno 45 stěrů z krčku děložního žen a 45 ejakulátů. M. hominis bylo prokázáno u 2 (4,4 %) žen a 3 (6,7 %) mužů. Frekvence výskytu U. urealyticum u obou pohlaví byla významně vyšší. U. urealyticum bylo potvrzeno u 19 (42,2 %) cervikálních stěrů a 9 (20 %) ejakulátů. Přítomnost uvedených bakteriálních druhů v ejakulátu byla spojena s negativním vlivem na hodnoty spermatických parametrů.

Abstract:

The aim of this thesis was to establish the incidence of Mycoplasma hominis and Ureaplasma urealyticum in the urogenital tract of men and women treated at the Centre of Assisted Reproduction SANUS Pardubice. The microorganisms were detected by species-specific polymerase chain reaction. At the same time the effect of Mycoplasma hominis and Ureaplasma urealyticum contamination on semen quality was determined. The results were then compared with similar studies. Overall, 45 cervical swabs and same number of ejaculates were examined. M. hominis was detected in 2 (4.4 %) cervical specimens and 3 (6.7 %) ejaculates. Prevalence of U. urealyticum in clinical samples taken from both sexes was significantly higher. U. urealyticum was confirmed in 19 (42.2 %) cervical swabs and 9 (20 %) ejaculates. The presence of these bacterial species in the ejaculate was associated with negative effect on sperm parameters.
