Head and neck cancer cell exosome isolation and characterization

Abstract:

Head and neck spinocellular cercinoma is an aggressive type of cancer and is one of the six most frequent causes of death by cancer. This bachelor´s thesis focuses on processes of autophagy and exosomal secretion in head and neck spinocellular carcinoma cells. We addressed the influence of bafilomycin, cannabidiol, doxorubicin and rapamycin on the autophagic activity of cells and on the extent of production of exosomes. We also optimized the method for isolation and elution of exosomes. To isolate the exosomes, we used immunoaffinity methods using magnetic beads and glycine buffer and repetitive rinsing with PBS solution with 0,1 % BSA to elute them. The extent of production of exosomal vesicles after applying the exogenic substances was monitored by Western blotting and the autophagic activity was monitored by using CYTO-ID and fluorescence microscopy. The result showed that by eluting the exosomes with glycine buffer exosomes of higher purity were obtained, whereas the method of repetitive rinsing showed to be less effective. Inhibitory effect of bafilomycin on autophagic processes was proved experimentally, whereas doxorubicin, rapamycin and especially cannabidiol showed activating effects. The effect of these substances on the production of exosomes was, however, not proven, because we did not manage to optimize the conditions for Western blotting.

Abstract:

Karcinom hlavy a krku je agresivní nádorové onemocnění a patří mezi šest nejčastějších příčin úmrtí na solidní nádory. Tato práce se zabývá zkoumáním procesů autofágie a exozomální sekrece u buněk odvozených ze spinocelulárního karcinomu v oblasti hlavy a krku. Sledovali jsme vliv bafilomycinu, kanabidiolu, doxorubicinu a rapamycinu na autofagickou aktivitu buněk a míru tvorby exozomálních vezikul. Zároveň jsme optimalizovali metodu izolace a eluce exozomů. K izolaci exozomů byly použity imunoafinitní zachytávací metody a k jejich eluci glycinový pufr a repetitivní oplachování roztokem PBS s 0,1% BSA. Míra tvorby exozomálních vezikul po působení exogenních látek byla sledovaná Western blotem a autofagická aktivita byla pozorovaná pomocí barvení CYTO-ID fluorescenčním mikroskopem. Bylo zjištěno, že elucí exozomů glycinovým pufrem byly získány exozomy ve vyšší čistotě v porovnání s metodou repetitivního oplachu. Experimentálně byl prokázaný inhibiční účinek bafilomycinu na autofagické procesy, doxorubicin, rapamycin a především kanabidiol naopak vykazovaly aktivační účinky. Vliv těchto látek na tvorbu exozomů se bohužel zjistit nepodařilo, protože se nepodařilo optimalizovat podmínky Western blottingu.
