The study of protein BRCA1-DNA interactions.

Abstract:

Produkt genu BRCA1 zastává funkci nádorového supresoru, jehož hlavní úlohou je snaha o zachování genomové integrity. Přítomnost jaderného fosfoproteinu BRCA1 byla jasně prokázána během opravy dvojřetězcových zlomů DNA, v kontrolních bodech buněčného cyklu, duplikace centrozomů a transkripce, během signalizace poškození DNA, růstové regulace a indukce apoptózy. Svou roli často plní skrz interakci s jinými proteiny, sám je však schopen i přímé vazby k DNA skrz určité úseky v dlouhé neuspořádané centrální oblasti. Pro přímou vazebnou interakci s DNA protein BRCA1 silně upřednostňuje větvené struktury DNA, což zřejmě souvisí s jeho rolí při reakci na poškození genetické informace. V případě vazby na lineární DNA je vazebně aktivní až od určité délky vázaného lineárního duplexu (300-500 bp). Fosfoprotein BRCA1 se k DNA váže převážně sekvenčně nespecificky, existuje však možnost, že dokáže vyhledávat cílovou sekvenci TTCTGTTG.

Abstract:

The product of the BRCA1 gene plays the role of a tumour suppressor of which the main task is to maintain genome integrity. The presence of the nuclear phosphoprotein BRCA1 has been demonstrated during the reparation of double-strand DNA breaks, in cell cycle checkpoints, centrosome duplication and transcription, in DNA damage signalling pathways, growth regulation and induction of apoptosis. BRCA1 often fulfils its function by interacting with others proteins, however it is capable of direct DNA binding of its own. BRCA1 displays a strong preference for branched DNA structures, which is apparently related to its role in the reaction to damage of genetic information. In the case of linear DNA binding, BRCA 1 actively binds only linear duplex longer than 300-500 bp. The BRCA1 phosphoprotein interacts with DNA mainly without DNA sequence specificity, though it is possible it can interact specifically with the target sequence TTCTGTTG.
