Decarboxylase Activity of Selected Bacteria in the Presence of Bacteriocins

Abstract:

Tato diplomová práce se zabývá sledováním produkce biogenních aminů bakteriemi Lactobacillus plantarum RIBM 2-89 a Lactobacillus curvatus subsp. curvatus T3 v závislosti na přídavku bakteriocinu nisinu. Teoretická část je zaměřena na bakterie mléč-ného kvašení, jejich charakteristiku, jednotlivé druhy a využití v potravinářství. Dále jsou zde popsány bakteriociny produkované těmito bakteriemi se zaměřením na nisin. Nakonec je věnována pozornost charakteristice, funkci a výskytu biogenních aminů. V praktické části byl proveden skríning 21 kmenů bakterií izolovaných z piva a ze sýru na jejich dekarboxylázovou aktivitu. Na základě získaných výsledků byly vybrány 2 kmeny vykazující nejvyšší dekarboxylázovou aktivitu. U těchto kmenů byla sledována produkce biogenních aminů (tyraminu, sperminu) v závislosti na vybraných faktorech (přídavek nisi-nu, teplota). Produkce biogenních aminů byla analyzována pomocí vysokoúčinné kapalino-vé chromatografie s UV/VIS detekcí po předchozí derivatizaci dansylchloridem. U obou kmenů byl obsah biogenních aminů vyšší během kultivace při 30 °C. S přídavkem nisinu o vyšší koncentraci došlo ke snížení obsahu biogenních aminů. Přídavek nisinu v různých časových intervalech neměl jednoznačný vliv na obsah biogenních aminů v bujónu.

Abstract:

This thesis deals with monitoring of biogenic amines production by bacteria Lactobacillus plantarum RIBM 2-89 and Lactobacillus curvatus subsp. curvatus T3 depending on the addition of bacteriocin nisin. The theoretical part is focused on lactic acid bacteria, their characteristics, species and use in food processing. Further there is description of the bacte-riocins produces by these bacteria, focusing on nisin. Finally, attention is beeing paid to the characteristics, function and the occurrence of biogenic amines in food. In practical part there was perfomed screening of 21 bacterial strains isolated from beer and cheese on their decarboxylase activity. On the basis of the obtained results have been selec-ted 2 strains with the highest decarboxylase activity. In these strains was monitored produ-ction of biogenic amines (tyramine, spermine) depending on selected factors (addition of nisin, temperature). Production of biogenic amines was analyzed by high performance liquid chromatography with UV/VIS detection after derivatization with dansylchloride. For both strains was the content of biogenic amines higher during cultivation at 30 °C. With the addition of higher concentration of nisin came to a reduction of the content of biogenic amines. The addition of nisin in different time intervals didn´t have a clear effect on the content of biogenic amines in medium.
