Bacterial Production of Biogenic Amines in Milk with Different Fat Content

Abstract:

Tato diplomová práce se zabývá stanovením obsahu biogenních aminů v mléce o různé tučnosti. Teoretická část je věnována charakteristice biogenních aminů a mikroorganismů s dekarboxylázovou aktivitou, dále stanovení biogenních aminů a popisu mléka. V praktické části byl v mléce v podmínkách in vitro sledován vliv několika faktorů (mléko o různé tučnosti bez úpravy a mléko s přídavkem aminokyselin, teplota a doba kultivace) působících na produkci 8 biogenních aminů (tryptaminu, fenyletylaminu, putrescinu, kadaverinu, histaminu, tyraminu, sperminu a spermidinu) testovanými kmeny bakterií. Vyprodukované biogenní aminy byly stanoveny pomocí vysokoúčinné kapalinové chromatografie s UV/VIS detekcí po předkolonové derivatizaci dansylchloridem. Všechny testované kmeny produkovaly putrescin a tyramin. Koncentrace dalších biogenních aminů se u jednotlivých kmenů lišila. Nejvyšší produkci biogenních aminů, s obsahem 97 mg/l prokázal v mléce plnotučném bez aminokyselin Staphylococcus thermolyticus DEPE22. Nejvyšší dekarboxylázová aktivita v mléce s přídavkem aminokyselin byla stanovena u Enterococcus faecalis CCM2665, u kterého byla detekována suma biogenních aminů 2728 mg/l, a to taktéž v mléce plnotučném. Lactobacillus curvatus AI-11 vykazoval nejnižší dekarboxylázovou aktivitu v mléce nízkotučném, zde hodnota vyprodukovaných biogenních aminů dosáhla pouze 3 mg/l. Nejnižší detekované množství biogenních aminů v mléce s přídavkem AMK, a to 21 mg/l, bylo dosaženo kmenem Lactobacillus curvatus T2 v mléce nízkotučném.

Abstract:

This thesis deals with the determination of biogenic amines in the milk of various fatness. The theoretical part is devoted to the characteristics of biogenic amines and microorganisms with decarboxylase activity, the determination of biogenic amines and description of milk. The practical part of the milk in vitro is influenced of several factors (milk with different fat content without modification and milk with added amino acids, temperature and duration of culturing) acting on the output 8 of biogenic amines (tryptamine, phenylethylamine, putrescine, cadaverine, histamine, tyramine, spermine and spermidine) tested strains. Biogenic amines produced were determined by high performance liquid chromatography with UV/VIS detection after precolumn derivatization with dansyl chloride. All test strains produced putrescine and tyramine. Concentrations of other biogenic amines for individual strains was different. Highest producing biogenic amines, was containing 97 mg/l, it was showed in the full-fat milk without AA by Staphylococcus thermolyticus DEPE22. Highest decarboxylase activity in milk with the addition aminoacids was determined by Enterococcus faecalis CCM2665 in which the detected amount of biogenic amines was 2728 mg/l, in the full-fat milk also. Lactobacillus curvatus AI-11 showed the lowest decarboxylase activity in the low-fat milk. The value of biogenic amines reached only 3 mg/l. The lowest detected amount of biogenic amines in the low-milk with the addition of AA was 21 mg/l achieved strain by Lactobacillus curvatus T2 in the low-fat milk.
