The ratio of glutathione disulfide to glutathione as a possible marker of oxidative stress in humans

Abstract:

V diplomové práci jsme ověřovali metodu měření celkového glutathionu (tGSH) a glutathiondisulfidu (GSSG) pomocí vysokoúčinné kapalinové chromatografie s fluorescenční detekcí. Vyšetřovali jsme také vhodná antikoagulační, alkylační, redukční a deproteinační činidla s ohledem na stabilitu a výtěţnost. Glutathion reaguje s o-ftaldialdehydem (OPA) za vzniku stabilního, vysoce fluoreskujícího tricyklického derivátu při pH 8, zatímco GSSG reaguje s OPA při pH 12. Při měření GSSG byl GSH blokován N-ethylmaleimidem, s nímţ vytváří GSH komplex. Separace probíhala na koloně Discovery C18 150 x 4 mm, 5 ?m. Mobilní fází byla směs ethanolu a 25 mmol/l Na2HPO4 (8:92, v/v) o pH 6,0. U skupiny dárců krve jsme stanovili referenční rozmezí celkového glutathionu a glutathiondisulfidu v plné krvi (9,35 ? 1,28) ?mol/g hemoglobinu (Hb) a (0,38 ? 0,06) ?mol/g Hb a plazmě (5,19 ? 1,23) ?mol/l a (0,36 ? 0,13) ?mol/l. Tato HPLC metoda je velmi citlivá a reprodukovatelná pro stanovení celkového glutathionu a glutathiondisulfidu v plné krvi a plazmě.

Abstract:

A method for the measurement of total glutathione (tGSH) and glutathione disulfide (GSSG) using an HPLC with fluorescence detection and investigation into the suitability of anticoagulant, alkylating, reducing and deproteinizing agents with regard to stability and recovery are described. Glutathione reacts with ortophthalaldehyde (OPA) to form a stable, highly fluorescent tricyclic derivate at pH 8.0, while GSSG reacts with OPA at pH 12.0. At measurement of GSSG, GSH was complexed to N-ethylmaleimide. For the separation, a reserve phase column Discovery C18, 150 mm x 4 mm, 5 ?m, was used. The mixture of ethanol and 25 mmol/L sodium hydrogenphosphate (8:92, v/v), pH 6.0, was used as a mobil phase. The preliminary reference ranges of total glutathione and glutathione disulfide in a group of blood donors are (9.35 ? 1.28) ?mol/g of hemoglobin (Hb) and (0.38 ? 0.06) ?mol/g Hb for whole blood, (5.19 ? 1.23) ?mol/L and (0.36 ? 0.13) ?mol/L for plasma. This assay is a highly sensitive and reproducible HPLC method for the determination of total glutathione and glutathione disulfide in human whole blood and plasma.
