Study of cisplatin derivatives effects on cancer cell

Abstract:

Platinová cytostatika patří mezi nejčastěji používaná protinádorová léčiva. Častý výskyt nežádoucích vedlejších účinků a vznik rezistence jsou však závažnými problémy limitujícími jejich použití. Cílem je proto zavedení nových derivátů, které by odstranily nežádoucí účinky, překonaly rezistenci a měly lepší protinádorovou aktivitu. Tato práce se zabývá studiem mechanismu účinku nového platinového derivátu LA-12. Díky struktuře platinového komplexu obsahujícího hydrofóbní ligand adamantylamin proniká LA-12 do buněk mnohem rychleji a účinněji než cisplatina, což přináší mnohem silnější protinádorové účinky i u buněk k cisplatině rezistentních. Ve vztahu k důležitému nádorovému supresoru, proteinu p53, jsme zjistili, že LA-12 je schopna účinně indukovat apoptózu i u buněk neobsahujících funkční p53 protein, tedy nezávisle na přítomnosti funkčního proteinu p53. Při studiu mechanismu působení LA-12 na molekulární úrovni jsme objevili, že LA 12 funguje jako účinný inhibitor molekulárního chaperonu Hsp90 a tento poznatek jsme potvrdili několika přístupy. Tyto jedinečné vlastnosti LA-12 tak otevírají možnosti pro navržení nových léčebných strategií v nádorové terapii.

Abstract:

Platinum cytostatics are effective and commonly used anticancer drugs. However, the therapy using cisplatin has large limitations including serious side effects and frequent emergence of intrinsic or acquired resistance. For this reason, vast efforts were committed to develop novel platinum-based complexes to overcome platinum resistance, to reduce cisplatin side effects and to introduce novel mechanisms of anti-cancer action. This work is focused on the study of mechanism of action of new platinum derivative LA-12. Thanks to the structure of the platinum complex containing hydrophobic ligand adamantylamine, LA-12 penetrates to the cells much faster and more effectively than cisplatin resulting in improved cytotoxicity in various cancer cell lines, including cisplatin resistant cells. In connection with important tumour suppressor p53, we found that LA-12 is capable of efficient induction of apoptosis also in the cells, which do not contain functional p53 protein and thus functions independently of functional p53. Studying the mode of action of LA-12 on a molecular level, we found, that LA-12 functions as an inhibitor of molecular chaperone Hsp90 and this finding was confirmed by several approaches. These unique properties of LA-12 therefore open possibilities to design new cancer treatment strategy.
