Factors Influencing the Ability of Probiotic Bacteria to Form Biogenic Amines

Abstract:

Tato práce byla zaměřena na studium metabolických drah biosyntézy putrescinu u vybraných kmenů rodu Lactobacillus a na podmínky ovlivňující produkci biogenních aminů probiotickým kmenem Lactobacillus rhamnosus. Metodou PCR bylo zjištěno, že hlavní cestou syntézy putrescinu u kmenů Lb. curvatus je agmatideaminázová dráha. Pomocí vysokoúčinné kapalinové chromatografie byl sledován vliv pH, koncentrace NaCl a koncentrace laktózy na dekarboxylázovou aktivitu kmene Lactobacillus rhamnosus. Za daných kultivačních podmínek byl nejvíce produkovaným biogenním aminem spermin. Detekované množství sperminu se pohybovala v rozmezí 17,9 ? 24,3 mg/l kultivačního média. Vliv pH byl nejvýraznější u produkce spermidinu a putrescinu. Hodnota pH 5,0 působila inhibičně na produkci spermidinu a také se výrazně snížila produkce putrescinu. V kultivačních médiích obsahující 2 % (w/v) NaCl bylo detekováno nejvyšší množství tyraminu (12,6 mg/l) a putrescinu (10,6 mg/l).

Abstract:

This work was focused on the study of metabolic pathways of putrescine biosynthesis by selected strains of Lactobacillus and the effect of selected factors on the production of biogenic amines with probiotic strain Lactobacillus rhamnosus. PCR detection showed that the major pathway of putrescine synthesis in strains Lb. curvatus is agmatideaminase pathway. Using high performance liquid chromatography, the influence of pH, concentration of NaCl and the concentration of lactose on decarboxylase activity Lactobacillus rhamnosus was tested. Under these conditions was the most produced biogenic amine spermine. The amount of spermine ranged from 17.9 to 24.3 mg/l culture medium. Effect of pH was most pronounced for the putrescine and spermidine production. PH 5.0 acted by inhibiting the production of spermidine and also significantly reduced the production of putrescine. The culture media containing 2% (w/v) NaCl were detected maximum amount of tyramine (12.6 mg/l) and putrescine (10.6 mg/l).
