Abstract:

Staphylococcus aureus je oportunním patogenem vyskytujícím se v části populace jako součást přirozené mikroflóry člověka. V současně době představuje největší problém jeho zvyšující se rezistence k mnoha druhům antibiotik. Svůj podíl na tomto fenoménu mají také kmeny S. aureus tvořící trpasličí kolonie, které představují pomalu rostoucí auxotrofní subpopulace bakterií s charakteristickými fenotypovými a patogenními znaky. Tyto kmeny slouží jako klidová stádia pro přečkání doby antibiotické terapie a jsou tedy zodpovědné za rekurentní chronické infekce u pacientů trpících zejména cystickou fibrózou. V poslední době se rozrůstá zájem o tuto problematiku, kdy je většina studií zaměřena na objasnění vzniku auxotrofních mutací a s nimi spojenou antibiotickou rezistenci. Tato diplomová práce je zaměřena naopak na charakterizací meticilin rezistentních trpasličích kmenů na molekulární úrovni. Současně je také věnována testování citlivosti těchto kmenů k bakteriofágům. Dále se práce zabývá stanovením dynamiky genomu chronologicky izolovaných trpasličích kmenů.

Abstract:

Staphylococcus aureus is an opportunistic pathogen occurring in the part of the population as a part of natural microflora of humans. At present, the biggest problem is its increasing resistance to many types of antibiotics. The contribution to this phenomenon is also connected to strains of S. aureus with small-colony variant phenotype, which represents slowly growing auxotrophic subpopulations of bacteria with typical phenotypic and pathogenic characteristics. These strains serve as dormant stages to survive the period of antibiotic therapy and because of that they are responsible for recurrent chronic infections in cystic fibrosis pacients. Recently, there has been a growing interest in this issue, when the majority of studies are focused on clarification of the emergence of auxotrophic mutations and connected antibiotic resistance. This diploma thesis is on the contrary focused on characterization of methicillin resistant small colony variants on molecular level. It is also devoted to testing the sensitivity of these strains to bacteriophages. The work deals with the determination of the genome dynamics from chronologically isolated small colony variants too.
