Centrosome aberrations in tumor cells of neurogenic origin

Abstract:

Centrosom má funkci mikrotubuly organizujícího centra zajišťující nukleaci a organizaci mikrotubulů v buňce. Každá buňka by měla mít během mitózy dva centrosomy, které vytvářejí bipolární mitotické vřeténko a tím zajišťují rovnoměrný přenos chromosomů do dceřiných buněk. Deregulace duplikace centrosomu vede k výskytu multipolárního mitotického vřeténka. Takové multipolární mitotické vřeténko je příčinou chybného přenosu chromosomů do dceřiných buněk, a to nakonec způsobuje aneuploidii, která je charakteristickým znakem nádorově transformovaných buněk. Diplomová práce je zaměřena na studium aberací centrosomů v buňkách vybraných nádorových buněčných linií neurogenního původu, zejména ve vztahu k expresi dvou N-terminálních izoforem proteinu p73 a k interakcím těchto izoforem s proteinem BubR1. Izoformy TAp73 a ∆Np73 vykazovaly převážně jadernou lokalizaci u třech buněčných linií. U většiny buněk byla také zjištěna cytoplazmatická lokalizace, kdy se protein vyskytoval ve specifické oblasti v blízkosti buněčného jádra. Z výsledků vyplývá, že izoforma ∆Np73 má vliv na počet centrosomů v buňce. Zatímco se exprese izoformy TAp73 neměnila, exprese izoformy ∆Np73 byla ve většině buněk s aberantním počtem centrosomů zvýšená. Dále byly popsány rozdíly v intracelulární lokalizaci BubR1 v závislosti na buněčném cyklu. Vztah mezi BubR1 a aberacemi centrosomů, resp. izoformami p73 však nebyl potvrzen. Tato práce byla podpořena grantem MUNI/C/0804/2011.

Abstract:

Centrosome has a function of a microtubule organizing center providing nucleation and organization of microtubules in the cell. Each cell should have two centrosomes during mitosis, which generate bipolar mitotic spindle and thereby ensure an even distribution of chromosomes into the daughter cells. A deregulation of centrosome duplication leads to the occurrence of a multipolar mitotic spindle. Such multipolar spindle may cause an irregular distribution of chromosomes into daughter cells, which is the hallmark of transformed cells. The diploma thesis is focused on the study of centrosome aberrations in selected tumor cell lines of neurogenic origin, in particular in relation to the expression of two N-terminal isoforms of p73 protein and interactions of these isoforms with BubR1 protein. The TAp73 and ΔNp73 isoforms showed predominantly nuclear localization in three cell lines. In these cell lines was also identified cytoplasmic localization of these isoforms in a specific region near the cell nucleus. The results show that ΔNp73 isoform affects the number of centrosomes in the cell. While the expression of TAp73 isoform was not changed, expression of ΔNp73 isoform was increased in most cells with aberrant number of centrosomes. Furthermore, the differences in the intracellular localization of BubR1 depending on the cell cycle were described. However, the relationship between BubR1 and centrosome aberrations, respectively p73 isoforms was not confirmed. This diploma thesis was supported by a MUNI/C/0804/2011 grant.
