The importance of PIWI-interacting RNA pathway in the glioblastoma stem cell biology

Abstract:

Glioblastom (GBM) je nejčastějším a velmi agresivním primárním nádorem mozku, u kterého se průměrné přežití pacientů pohybuje pouze okolo 12 až 15 měsíců od stanovení diagnózy. To je způsobeno jeho lokalizací a extrémní invazivitou, která komplikuje chirurgickou resekci a je hlavní příčinou pozdějšího návratu onemocnění. Subpopulace takzvaných nádorový kmenový buněk také velkou měrou přispívá k heterogenitě a agresivitě onemocnění. Tyto buňky jsou pravděpodobně zodpovědné za vznik a rozvoj nádoru, zisk rezistence vůči standardní chemo a radioterapii, čímž se zásadně podílejí na relapsu onemocnění. Z těchto důvodů je jejich identifikace a případné cílení potenciálně silným nástrojem v terapii GBM. Jedním z možných markerů těchto buněk by tak mohly být i krátké nekódující RNA, konkrétně PIWI-interagující RNA. Tyto 24-35 nukleotidů dlouhé molekuly jsou zodpovědné za kontrolu aktivity transpozonů, čímž zajišťují stabilitu genomu, byla však popsána i řada dalších biologických rolí ve kterých hrají stěžejní funkci. Jejich dysregulace byla také pozorována u řady nádorů, včetně GBM. Cílem této diplomové práce je porovnat expresi PIWI-interagující RNA mezi nádorovými kmenovými buňkami glioblastomu a diferencovanými buňkami glioblastomu a identifikovat signifikantně dysregulované molekuly. U molekuly piR-9491 bylo dále provedeno několik in vitro funkčních testů ve snaze otestovat její funkční zapojení do regulace specifických biologických vlastností nádorových kmenových buněk glioblastomu.

Abstract:

Glioblastoma (GBM) is the most common and a highly aggressive primary brain tumour. The mean life expectancy of patients suffering with GBM is only around 12 to 15 months from the initial diagnosis. This is due to the localization and extreme invasivity of GBM which complicates its chirurgical resection and is the main cause for the tumour recurrence. Subpopulation of so-called cancer stem cells which are present in the tumour tissue also contributes to the heterogeneity and aggressivity of this disease. Cancer stem cells are probably responsible for the initiation and growth of the tumour and for its resistance against standard treatment thereby contributing to the relapse of the tumour. For this reasons identification and targeting of cancer stem cells could be a powerful tool used in the therapy of GBM. One of the possible markers for the cancer stem cell population could be short non-coding RNAs, specifically PIWI-interacting RNAs. These 24-35 nucleotide long molecules are mainly responsible for the suppression of transposon activity thereby ensuring the stability of genome. In recent years, other biological functions in which piRNA play a crucial part were documented. Dysregulation of piRNA expression was observed in a number of tumours, including GBM. The goal of this diploma thesis is to compare the expression of piRNA between differentiated cancer cells and cancer stem cells in glioblastoma and identify significantly dysregulated piRNA molecules. Further, in vitro experiments were performed on one of the dysregulated molecules, piR-9491 in order to elucidate its role in regulation of biological characteristics of glioblastoma cancer stem cells.
