Mechanisms of RNA quality control in the nucleus

Abstract:

V předkládané práci sleduji několik aspektů kontroly kvality RNA v kvasinkách a v savčích buňkách. První část dizertace se zabývá mechanizmy fungujícími v jádře kvasinky Saccharomyces cerevisiae, zejména proteinovými komplexy, které se účastní terminace transkripce a zpracování nekódujících RNA pomocí RNA polymerázy II. Charakterizuji RNA-vazebné vlastnosti komplexu NNS (Nrd1, Nab3 a Sen1) a molekulové mechanizmy aktivace NNS RNA polymerázou II. V druhé části práce se soustředím na lidský protějšek komplexu NNS, a to komplex NEXT (Nuclear Exosome Targeting), a na kontrolu kvality RNA a na degradaci v cytoplazmě savčích buněk. Popisuji RNA-vazebné vlastnosti podjednotky RBM7 komplexu NEXT a poukazuji na její úlohu ve zpracování a degradaci snRNA. Třetí část usiluje o charakterizaci role uridylovaného 3' konce RNA. Při hledání faktorů působících na netemplátovém oligo(U) 3' konci jsme identifikovali 3'-exonukleázu DIS3L2. Naše experimenty ukazují, že DIS3L2 je nukleáza, která selektivně štěpí uridylované prekurzory rodiny let-7 patřící mezi miRNA.

Abstract:

In this thesis I am providing an insight into several aspects of RNA quality control in yeast and in mammalian cells. The first part of the dissertation concerns mechanisms operating in the nucleus of budding yeast Saccharomyces cerevisiae, in particular protein complexes acting during transcription termination and processing of noncoding RNAs by RNA polymerase II. I was involved in the characterization of the RNA-binding properties of the NNS (Nrd1, Nab3 and Sen1) complex and the molecular basis of NNS recruitment by RNA polymerase II. The second part of the thesis work is focused on the NNS-like complex in human cells, called NEXT (Nuclear Exosome Targeting) and on the RNA quality control and degradation in mammalian cytoplasm. Briefly, we have characterized the RNA-binding properties of the RBM7 subunit of the NEXT and point to its role in snRNA processing and degradation. The third part describes our attempts to characterize the role of 3’terminal RNA uridylation. Our search for factors acting on the oligo(U) 3‘ nontemplated termini resulted in the identification of the cytoplasmic 3’ to 5’ exonuclease DIS3L2. Our experiments showed, that DIS3L2 is a nuclease that targets uridylated precursors of the let-7 family of miRNAs.
